Total Synthesis of Atorvastatin

摘要 阿托伐他汀钙(HMG-Co A还原酶选择性抑制剂)作为全世界应用最广泛的降胆固醇药物之一,有关其合成方法的研究吸引了化学家们的广泛关注.以廉价易得的(R)-2-氯甲基代环氧乙烷为原料,经过13步反应,以9%的总收率成功地合成了阿托伐他汀,为相应的仿制药的生产提供了新的路线选择. Atorvastatin calcium (HMG-CoA reductase inhibitor) is the most widely used cholesterol lowering drugs. Its synthesis has attracted broad attention of synthetic chemistry community. In this study, a new strategy has been developed toward the synthesis of atorvastatin in 13 steps and 9% overall yield.
