摘要
研究了头孢米诺与维多利亚蓝B之间的显色反应,确定了最佳反应条件,建立了简便、快速、灵敏测定头孢米诺的分光光度法。在pH=7.05的Tris-HCl缓冲介质中,适量的头孢米诺与维多利亚蓝B反应,生成具有明显正、负吸收峰的配合物。产物的最大正吸收波长为630nm,最大负吸收波长为424nm,表观摩尔吸光系数(ε)分别为4.20×104 L/(mol·cm)(正吸收)和1.69×104 L/(mol·cm)(负吸收)。头孢米诺质量浓度在0-5.2mg/L范围内服从比耳定律。该方法用于测定头孢米诺钠药物及人体尿样中头孢米诺的含量,结果满意。
The chromogenic reaction between cefminox and victoria blue B was investigated by spectrophotometry.The experimental optimum conditions were confirmed.A simple,rapid and highly sensitive method for determination of cefminox was developed.In Tris-HCl buffer medium of pH =7.05,an appropriate amount of cefminox reacted with victoria blue B to form a complex with obvious positive absorption peak and negative absorption peak.The maximum positive absorption wavelength was at 630 nm and maximum negative absorption wavelength was at 424 nm.The apparent molar absorptivity(ε)were 4.20×104(positive absorption)and 1.69×104(negative absorption)L/(mol·cm),respectively.Beer's law was obeyed in the range of 0-5.2mg/L.The method has been applied to the determination of cefminox in the commercially available cefminox sodium medicament and human urine with satisfactory results.
