摘要
目的研究通窍活血汤(川芎、赤芍、桃仁、红花等)对血管性痴呆大鼠学习记忆和血液流变学的影响。方法通过改良的双侧颈总动脉结扎,建立拟血管性痴呆大鼠模型。给药31 d后,Morris水迷宫实验对大鼠的学习记忆能力进行检测。大鼠腹主动脉取血后,通过血液流变仪检测大鼠的在低、中、高切全血黏度和血浆黏度,血液分析仪检测大鼠的红细胞变形指数和红细胞聚集指数。结果与模型组相比,通窍活血汤高、中、低剂量能显著减少血管性痴呆大鼠逃避潜伏期和找到并爬上平台的总路程(P<0.01);与假手术组相比,模型组低切、中切下的全血黏度、血浆黏度、红细胞变形指数及红细胞聚集指数明显提高(P<0.05,P<0.01);与模型对照组相比,通窍活血汤高、中、低剂量组大鼠的红细胞变形指数、红细胞聚集指数明显降低(P<0.05),低切、中切、全血黏度及血浆黏度明显降低(P<0.01)。结论通窍活血汤能明显改善血管性痴呆大鼠学习和记忆能力,其机制可能与其改善血液流变学指标有关。
AIM To study the influence of Tongqiao Huoxue Decoction( TQHXD,Chuanxiong Rhizoma,Paeoniae Radix rubra,Persicae Semen,Carthami Flos,etc.) on the memory and learning ability and the efficacy of hemorheology on vascular dementia( VD) rats. METHODS VD rat model was established by optimized bilateral carotid artery ligation. The MORRIS water maze experiment was adopted to evaluate the ethology of the VD rats. Blood was drawn from abdominal aorta. Their whole blood viscosity and plasma viscosity were detected at low,middle and high shear situations in VD rats by Rheology instrument. The hematocrit,the erythrocyte aggregation index and the erythrocyte rigidity index in VD rats were analyzed by hematorogy analyser. RESULTS Compared with the model group,high,middle and low dose groups of TQHXD significantly reduced the time of escape latency and the distance of climbing on the platform( P 〈0. 01). Compared with the sham group,the model group markedly increased in whole blood viscosity at low and middle shear situations,plasma viscosity,erythrocyte aggregation index and erythrocyte deformation index( P 〈0. 01,P 〈0. 05) in those model groups also increased.Compared with the model group,high,middle and low dose groups of TQHXD decreased the whole blood viscosity and plasma viscosity at low and middle shear situations( P 〈0. 05). Erythrocyte aggregation index and erythrocyte deformation index also obviously decreased( P 〈0. 01). CONCLUSION TQHXD can significantly improve the memory and learning ability of VD rats. Their mechanism of action might be related to its effects on the hemorheology.
