淡豆豉和黑豆乙醇提取物的促成骨细胞增殖活性及HPLC-MS分析 被引量:8

摘要 目的比较分析淡豆豉和黑豆乙醇提取物促大鼠颅骨成骨细胞增殖活性及二者的化学成分。方法采用MTT法对淡豆豉和黑豆的乙醇提取物进行促大鼠颅骨成骨细胞增殖活性的研究,并通过高效液相色谱-质谱联用技术对二者化学成分进行比较分析。结果淡豆豉和黑豆的乙醇提取物对大鼠成骨细胞的促增殖率分别可达到36.5%和28.2%,其主要有效成分为异黄酮类化合物,经液质联用分析共鉴定出15个黄酮类成分。结论黑豆发酵成淡豆豉后的成分变化主要体现在由苷类成分脱糖转化为苷元,含有量显著增加的大豆苷元和染料木素两种苷元主要由丙二酰大豆苷(大豆苷元-G-M)和丙二酰染料木苷(染料木素-G-M)脱丙二酰基和葡萄糖转化而来。 AIM To investigate the proliferative activity of ethanolic extract from black soybean( Sojae Semen Nigrum) and fermented soybean( Sojae Semen Praeparatum) on rat osteoblasts and HPLC-DAD-ESI-MS / MS analysis. METHODS The proliferation test was performed by using rat osteoblasts with MTT assays in vitro.Qualitative analysis was performed by HPLC-DAD-MS to analyze and compare major active components in both alcoholic extracts. RESULTS Ethanolic extracts of black and fermented soybean were demonstrated to show proliferative activity. Fifteen flavonoids were tentatively characterized based on UV and mass spectral analysis. CONCLUSION The experimental data show that when black soybean ferments into fermented soybean,heterosides undergoes the desugarization and converts into aglycon,which increases the contents of daidein and genistein by means of the demalony and deglucose of malonyldaidzin and malonylgenistin.
