华蟾素抑制人胃癌BGC-823细胞体外侵袭、迁移的机理 被引量:10

摘要 目的探讨华蟾素(Cinobufacin)体外抑制人胃癌BGC-823细胞侵袭的作用及其机制。方法以0.156 mg/L和0.313 mg/L华蟾素体外作用人胃癌BGC-823细胞,采用MTT法检测华蟾素对BCG-823细胞生长的抑制效果,Transwell小室法检测细胞的迁移和侵袭能力,Western blot法检测BGC-823细胞中NF-κB p56、MMP-9蛋白的表达量。结果华蟾素可浓度依赖性抑制体外BGC-823细胞增殖、迁移和侵袭,同时下调BGC-823细胞中NF-κB和MMP-9蛋白表达。结论华蟾素体外可抑制胃癌的转移,该作用可能通过下调NF-κB、MMP-9的表达相关。 AIM To investigate the anti-invasive effect of Cinobufacin on the human gastric carcinoma BCG-823 cells and its mechanism in vitro. METHODS BCG-823 cells were treated by 0. 156 mg / L and0. 313 mg / L Cinobufacin. Their inhibitory action was measured by MTT assay. The migration and invasive ability of BCG-823 cells were measured by Transwell assay. Western blot analysis was performed on NF-κB p56,MMP-9 in BCG-823 cells. RESULTS Cinobufacin significantly inhibited the growth,migration and invasive ability of BCG-823 cells in a dose-dependent manner. The expressions of NF-κB and MMP-9 were suppressed and the MMP-9 / TIMP-1 ratio reduced after the treatment with Cinobufacin. CONCLUSIONS Cinobufacin can inhibit the migration and invasion of BCG-823 cells probably by down-regulating the expression of NF-κB and MMP-9.
