中药枣仁宁神胶囊镇静催眠作用的实验研究

The study on the sedation hypnosis effect of Zaoren Ningshen capsules

目的观察枣仁宁神胶囊的镇静催眠作用。方法将50只小鼠分为5组,每组10只,分别为空白对照组、地西泮阳性对照药组、枣仁宁神胶囊高、中、低剂量组。分别给予等体积蒸馏水、0.002 g·kg-1地西泮、0.5 g·kg-1枣仁宁神胶囊、1.00 g·kg-1枣仁宁神胶囊、2.00 g·kg-1枣仁宁神胶囊,灌胃给药,测定小鼠自主活动和戊巴比妥钠催眠剂量小鼠入睡潜伏期和睡眠时间的变化。结果小鼠活动次数给药各组有减少趋势,但差异无统计学意义;站立次数地西泮组显著低于空白对照组(P<0.05);枣仁宁神胶囊中、高剂量组显著低于空白对照组(P<0.05)。给药各组小鼠入睡潜伏期无明显变化;持续睡眠时间地西泮组显著高于空白对照组(P<0.05);枣仁宁神胶囊各剂量组均显著高于空白对照组,其中枣仁宁神胶囊中、高剂量组更显著(P<0.05)。结论枣仁宁神胶囊明显减少小鼠自主活动和延长睡眠时间,具有镇静催眠作用。

Objective To observe the effects of sedation hypnosis of Zaoren Ningshen capsule. Methods 50 mice were divided into five groups averagely ： blank control group, diazepam positive control group, high - dose group of Zaoren Ningshen capsules, middle dose group of Zaoren Ningshen capsules, and low - dose group of Zaoren Ningshen capsules. The mice in blank control group were given the same volume of distilled water by gavage;The mice in diazepam positive control group were given 0.002g/kg diazepam by garage;The mice in high -dose group were given 2.00 g kg-1 Zaoren Ningshen capsule by gavage;The mice in middle dose group were given 1.00 g kg-1 Zaoren Ningshen capsules by gavage;The mice in low - dose group were given 0.5 g kg-1 Zaoren Ningshen capsules by ga- vage. The Zaoren Ningshen capsule＇ s effects on latency sleep time and instrument independent activities by giving sodium pentobarbital hypnotic sleep and dosage were observed. Results It was found that the movement frequency of mice in each drug administration group has a declining trend,but there was no significant difference;The stand -up times of mice in diazepam positive control group were less than the blank control group, and there was significant difference （P 〈 0.05 ） ; The stand - up times of mice in high - dose and middle dose group of Zaoren Ningshen capsule were less than blank control group, and there was significant difference （P 〈 0.05）. There is no obvious difference on sleep latency between mice in each drug administration group and blank control group; Sleep duration of mice in the diazepam positive control group was longer compared with the blank control group （ P 〈 0.05 ） ; Sleep duration of mice in each dose group of Zaoren Ningshen capsule was longer compared with the blank control group with significance in high and middle dose groups （ P 〈 0.05）. Conclusion Zaoren Ningshen capsule has the effect of reducing independent activities and extending the role of sleep time, namely Zaoren Ningshen capsule has strong hypnotic effects of sedation.
