摘要
目的研制与临床证候相吻合的切实可行的溃疡性结肠炎(脾肾阳虚证)的大鼠模型。方法将SD实验大鼠按随机数字表法随机分为4组:正常对照组、脾肾阳虚组、溃疡性结肠炎组(溃结组)、脾肾阳虚溃结组(病证结合组),正常对照组无任何干预措施;脾肾阳虚组先以番泻叶灌胃,然后用冰水湿润皮毛后再用电风扇将其皮毛吹干;溃结组以三硝基苯磺酸钠(TNBS)灌肠;病证结合组先以脾肾阳虚组的方法处理,连续处理4周后,再予TNBS灌肠。观察大鼠一般症状体征、中医证候、结肠组织的改变、血清大鼠游离三碘甲状腺原氨酸(FT3)、大鼠游离四碘甲状腺原氨酸(FT4)、超氧化物歧化酶(SOD)、丙二醛(MDA)等指标。结果病证结合组、脾肾阳虚组大鼠的中医证候评分、血清FT3、FT4、SOD、MDA等指标与正常对照组、溃结组相比,差异有统计学意义(P<0.05);病证结合组、溃结组结肠肉眼观及结肠黏膜损伤程度与正常对照组、脾肾阳虚组相比,差异具有统计学意义(P<0.05)。结论先以番泻叶灌胃及冰水湿身风干法处理大鼠,再用TNBS灌肠,从而研制出的溃疡性结肠炎(脾肾阳虚证)大鼠模型,对临床有实践意义。
Objective: To investigate establishment of the rat model with ulcerative colitis of Spleen-kidney yang deficiency type based on ulcerative colitis model. Methods: All the SD rats were divided randomly into 4 groups, namely the normal control (NC), the Spleen-kidney Yang Deficiency (SKYD), the ulcerative colitis (UC), and the SKYD with ulcerative colitis (SKYDUC). The rats in NC group had no intervention measures,in UC clystered by TNBS,in SKYD gavaged with folium sennae solution and washed with ice water, then dried their furs with electric fan,and in SKYDUC managed firstly with the same method as SKYD for four weeks,then elystered by TNBS. The following items were observed: the general symptoms and signs,the changes of syndrome of tradi- tional Chinese medicine,colon tissue and serum indicators such as FT3,FT4,MDA,SOD. Results: Compared with the NC and UC group,the differences of the TCM syndrome score,serum FT3,NF4,SOD and MDA in the SKYDUC and SKYD group were statistically significant (P〈 0.05). Compared with the NC and SKYD group,the difference of eye view of colon and colonic mucosa damage in SKYDUC and UC group was statistically significant (P〈 0.05 ). Conclusion: The method used to induce the model in SKYDCB is feasible in practice.
