摘要
目的探讨非转移乳腺癌患者外周血中游离无细胞的DNA(cfD NA)中ErbB-2基因启动子区5’CpG甲基化状态及其与肿瘤组织中Her-2表达的关系。方法分离非转移乳腺癌患者外周血中的cfD NA,利用重亚硫酸盐测序法(BSP)对分离出的cfD NA及外周血细胞DNA的ErbB-2基因启动子区进行5’CpG甲基化分析,免疫组化及FISH方法检测肿瘤组织中Her-2的表达情况。结果 ErbB-2基因启动子区5’CpG的甲基化只在乳腺癌患者的血浆cfD NA中检测到(19/40),在其对应的外周血血细胞中未发现;乳腺癌患者外周血cfD NA的ErbB-2基因启动子区5’CpG甲基化和其肿瘤组织Her-2的表达成负相关。结论乳腺癌患者外周血cfD NA ErbB-2启动子区5’CpG的甲基化状态与肿瘤组织Her-2表达呈负相关。提示外周血cfD NA ErbB-2启动子区5’CpG甲基化状态能从某种程度上反映乳腺癌患者肿瘤组织Her-2表达情况,有可能作为乳腺癌Her-2状态观察的新窗口,可用于帮助乳腺癌分子分型判断,是否能作为乳腺癌治疗的新靶点,并作为Her-2阳性患者早期诊断的指标,尚有待于进一步研究。
Objective To investigate the relationship of methylation of ErbB-2 gene 5' CpG of cfDNA in peripheral blood of non-metastatic breast cancer patients with expression of Her-2 in tumor tissue. Methods Isolate cell-free DNA in peripheral blood of non-metastatic breast cancer patients, analysis methylation of 5' CpG in ErbB-2 gene promoter using bisulfite-sequencing PCR ( BSP), detect the expression of Her-2 using immuno- histochemistry and FISH. Results Methylation of ErbB-2 gene promoter was only detected in plasma cfDNA of breast cancer patients (19/40), not found in the corresponding peripheral blood cells; Methylation of ErbB-2 gene 5' CpG of cfDNA in peripheral blood of breast cancer patients was negatively correlated with the expression of Her-2 in tumor tissue. Conclusions Methylation of ErbB-2 gene 5' CpG in peripheral blood of breast cancer patients was negatively correlated with the expression of Her-2 in tumor tissue. We can speculate that the methylation of ErbB-2 gene 5' CpG in peripheral blood of breast cancer patients can reflect the expression of Her-2 breast cancer tissues to some extent, which may be used as a new method to observe Her-2 status of breast cancer, and which also can be used to help determine the molecular typing of breast cancer, and used as a new target for breast cancer treatment, and as an early diagnosis indicator of Her-2 positive patients still needs further study.
