基于生理生化差异的黄菠萝雌雄株鉴别技术研究

摘要 [目的]研究黄菠萝幼苗早期雌雄鉴定技术。[方法]采用形态学、化学试剂法、保护酶测定方法,对比成熟黄菠萝雌雄株叶片差异。[结果]结果表明,黄菠萝叶片形态学方法和保护酶测定方法均不能作为鉴定雌雄株的方法(P<0.05);甲基红溶液染色的雄性植株提取液呈现棕红色,雌性植株提取液为橘红色;BTB法雌雄株提取液颜色差异明显,雄性植株提取液呈深蓝色,雌性植株提取液为棕黄色,吸收光谱值差异显著(P<0.01)。TTC法在485 nm下测定雌雄株提取液存在显著差异(P<0.01)。斐林试剂的显色反应和雌雄株提取液吸收光谱值存在差异(0.01<P<0.05)。[结论]试验表明甲基红法、BTB法、TTC法和斐林试剂法均可作为黄菠萝雌雄株鉴定的方法。 [ Objective ] The early gender identification techniques for Phellodendron amurense were studied. [ Method ] Using morphology, chemical reagent method , physiological and biochemical method to stain the extract of P. amurense ,while to get the UV spectrophotometer value, by the difference color of extract and spectral value of P. amurense to infer the gender of sapling. [ Result ] The results showed that the morphology and chemical reagent methods cannot identify the gender of P. amurense. The chromogenic reaction was significant differences male brown-red rendering, female orange. The differences male plants extract color by BTB method were significant, the male plant extract color dark blue, female brown, absorption spectra values were significantly different (P 〈 0.01 ). TTC method measured at 485 nm male and female plants were significantly different (P 〈 0.01 ) extract. Fehling's solution and the color reaction of male and female plants extract value differences in the absorption spectrum (0.01 〈 P 〈 0.05 ). [ Conclusion ] It showed that methyl red method, BTB method, TTC method and Fehling's solution methods can be identified gender of P. amurense.
