恩施土家族胃溃疡患者MIF-173G/C基因多态性表达

Expression of MIF-173G/C gene polymorphism in gastric ulcer patients from Enshi Tujia nationality

目的探讨恩施土家族胃溃疡患者巨噬细胞移动抑制因子(migration inhibitory factor,MIF)表达水平,分析MIF-173G/C多态性与胃溃疡发病相关性。方法对2013年2月-2014年2月恩施土家族苗族自治州中心医院收治的145例土家族胃溃疡患者及120名土家族健康体检者(对照组),应用SNa Pshot SNP分型技术确定MIF-173G/C等位基因型,ELISA法检测血清中MIF水平。结果恩施土家族胃溃疡患者MIF-173G/C的3种基因型频率分别为GG:0.428、GC:0.400、CC:0.172;对照组3种基因型频率分别GG:0.617、GC:0.317、CC:0.067。MIF-173G/C基因型在两组间差异有统计学意义(χ2=12.79,P=0.016)。胃溃疡组MIF水平明显高于对照组[(823.25±61.45)pg/ml vs(345.49±56.96)pg/ml,P=0.001];不同MIF-173G/C基因表型的胃溃疡患者血清中MIF含量差异有统计学意义(F=28.60,P=0.041)。结论 MIF-173G/C的基因多态性直接影响着MIF分泌,可能与胃溃疡的发病及病情相关。

Objective To analyze the serum level of macrophage migration inhibitory factor （MIF） in gastric ulcer pa- tients from Enshi Tujia nationality and evaluate the potential relationship between gastric ulcer and 173G/C polymorphism. Methods A total of 145 gastric ulcer patients who selected from Sep. 2013 to Feb. 2014 and a matched control group of 120 healthy volunteers were included. MIF-173 G/C genotypes were detected by SNaPshot SNP technique. Serum MIF lev- el was detected by ELISA. Results The frequencies of the three genotypes between control group and gastric ulcer group were significantly different （χ2 = 12.79, P = 0. 016）. The concentration of the serum MIF in gastric ulcer group was higher than that in control group [ （823.25 ± 61.45 ） pg/ml vs （345.49 ± 56.96） pg/ml, P = 0. 001 ]. The serum MIF concentra- tions among three genotypes were also significantly different （F = 28.60, P = 0. 041 ）. Conclusion MIF production in gastric ulcer patients is influenced by MIF173G/C gene polymorphism, which suggest that MIF173G/C gene polymorphism may play a role in the pathogenesis of gastric ulcer patients from Enshi Tujia nationality.
