瑞舒伐他汀对脑缺血再灌注损伤大鼠神经细胞凋亡及凋亡诱导因子表达的影响 被引量:6

摘要 目的观察瑞舒伐他汀对大鼠脑缺血再灌注损伤神经细胞凋亡及凋亡诱导因子(AIF)表达的影响,探讨瑞舒伐他汀的神经保护机制。方法选择健康雄性Wistar大鼠72只,随机分为假手术组、模型组(缺血再灌注损伤)和瑞舒伐他汀组,每组24只,每组又分为脑缺血再灌注后12、24、72h3个时间点,剔除造模过程中死亡的大鼠,每个时间点随机选取6只大鼠。分别取大鼠脑组织,检测神经细胞凋亡数和AIF蛋白阳性细胞数。结果模型组和瑞舒伐他汀组大鼠再灌注12、24、72h神经细胞凋亡数和AIF蛋白阳性细胞数表达较假手术组明显增加,而瑞舒伐他汀组神经细胞凋亡数[(47.12±6.15)个vs(67.31±20.12)个,(34.94±8.47)个vs(62.03±8.71)个,(24.02±7.02)个vs(56.63±6.72)个]和AIF蛋白阳性细胞数[(31.01±10.22)个vs(45.12±8.54)个,(22.93±6.97)个vs(37.34±8.38)个,(15.92±6.01)个vs(30.71±4.86)个]较模型组明显降低(P<0.01)。结论瑞舒伐他汀对脑缺血再灌注损伤有神经保护作用,其作用机制可能与抑制AIF蛋白、对抗细胞凋亡有关。 Objective To study the effect of rosuvastatin on apoptosis of nerve cells and expression of apoptosis inducing factor (AIF) in rats following cerebral I/R injury. Methods Seventy-two healthy male Wistar rats were randomly divided into sham operation group,model group(I/R in- jury)and rosuvastatin treatment group (24 in each group). The animals in each group were further divided into 12,24,and 72 h cerebral I/R groups (8 in each group). Rat brain tissue samples were taken to calculate the number of apoptotic nerve cells and to detect the expression of AIF-positive cells. Results The number of apoptotic nerve cells was significantly greater and the expression level of AIF-positive cells was significantly higher in model group and rosuvastatin treatment group than in sham operation group while the number of apoptotic nerve cells was significantly smaller and the expression level of AIF-positive cells was significantly lower in rosuvastatin treat- ment group than in model group at 12,24,72 h after reperfusion (P〈0.01). Conclusion Rosuv- astatin can protect rats against cerebral I/R injury by inhibiting the expression of AIF-positive cells and apoptosis of nerve cells.
