摘要
本研究通过基于DNA条形码的分子鉴定方法对广东省广州市白云山风景区一株马勃菌进行精准鉴定。以野外采集的子实体提取的基因组为模板,扩增ITS和EF1α片段,并进行测序及系统发育分析。获得了600 bp左右的ITS片段和500 bp左右的EF1α片段,经分子系统学分析表明该硬皮马勃菌为Scleroderma sinnamariense,从而对该真菌进行了基于DNA条形码的精准鉴定。
The aim of this study is to identify a Scleroderma sp. using DNA barcoding. ITS and EF1α genes were PCR am- plified and sequenced. Further,molecular phylogeny based on ITS and EF1α confirmed the genetic position of this fungus. About 600 bp of ITS fragment and about 500 bp of EF1α fragments were obtained. The molecular phylogenetic analysis indi- cated that the fungus was Scleroderma sinnamariense. The results provided an exact identification method for this macro-fun- gus by DNA barcoding.
