摘要
目的:观察桂枝茯苓汤对乳腺癌细胞株MCF-7的凋亡相关蛋白和细胞周期的影响。方法:将浓度为300μg/m L、900μg/m L的桂枝茯苓汤分别作用MCF-7细胞24 h、48 h后,采用四甲基偶氮唑盐法(MTT)法检测细胞抑制率,流式细胞仪方法检测MCF-7细胞周期各时向DNA含量的变化,细胞免疫化学方法检测线粒体凋亡通路相关蛋白Bcl-2、Bax、Caspase-3/9和Bid的表达情况。结果:与空白对照组比较,桂枝茯苓汤各浓度组细胞抑制率明显升高(P<0.05),其中G0/G1期细胞DNA含量明显升高(P<0.05),桂枝茯苓汤可以降低Bcl-2蛋白的表达,同时可以促进Bax、Caspase-3/9和Bid蛋白的表达。结论:桂枝茯苓汤能体外诱导乳腺癌细胞凋亡,其机制可能与线粒体凋亡通路有关。
Objective: To observe the effect of Guizhifuling decoction on the apoptosis related protein of breast cancer cell lines and cell cycle. Methods: The MCF-7 cells were cultured in the Guizhifuling decoction with the concentration is 300 and 900 μg·mL^-1 with 24 and 48 hours, then the cells were collected and use the MTr to test the inhibitory rate of MCF-7.The flow cytometry was used to detect the DNA content variation of each cell cycle. The immunocytochemistry method was used to study the effects of Guizhifuling decoction on the indexes of the induced MCF-7 including Bcl-2, Bax, Caspase-3/9 and Bid. Results: Compared with control group, the inhibitory rate of cell increased significantly with different concentration groups of Guizhifuling decoction (P〈0.05). The content of DNA in G0/G1 cells increased significantly (P〈0.05). Guizhifuling decoction can reduce the expression of Bcl-2 protein and promote the expression of Bax, Caspase-3/9 and Bid protein. Conclusion: Guizhifuling decoction could induce the apoptosis of breast cancer cells in vitro; the mechanism may be associated with the mitochondrial apoptotic pathway.
