摘要
目的:采用简易悬吊术制备大鼠和家兔下肢深静脉血栓模型,同时将大鼠模型和家兔模型做一比较,探讨简易悬吊术是否为一种简便、适合动态研究深静脉血栓的造模方法,大鼠和家兔哪一种动物更适合该造模方法。方法:选用36只体重为(250±20)g的SD大鼠和42只体重为(2.5±0.5)kg的清洁级家兔,家兔按随机数字表法分为2组:动物模型组和假手术组。大鼠作为对照组。动物模型组及对照组均采用简易悬吊术制备动物模型。造模后观察不同时点动物模型血管内膜及血栓不同时期的病理变化。大鼠模型和家兔模型做比较并与假手术组做观察比较。结果:采用简易悬吊术制作模型后,大鼠模型和家兔模模型动物下腔静脉内不仅均形成稳定的血栓,而且形成血栓的动物存活率比较高,符合人体静脉血栓形成急性期的病理演变。家兔模型比大鼠模型造模操作更简单。结论:简易悬吊术可以成功制作适合动态研究药物对深静脉血栓形成(DVT)作用机制的家兔或大鼠深静脉血栓模型,家兔更适应于该方法造模。
Objective: Using simple suspension for preparation of rabbit and rat model of deep vein thrombosis. At the same time, compared the model rats with rabbits ,explore simple suspension is a kind of simple and suitable for dynamic study of deep vein thrombosis (DVT) modeling method, rats and rabbits which animals is more suitable for the modeling method. Methods: Selection of 36 weighing (250a:20) grams of SD rats and 42 weighing (2.5+0.5)grams of cleaning stage rabbits .The rabbits were divided into animal model group and sham operation group with random digits table method. Rats served as control group. The animal model group and the control group were prepared by simple suspension operation. To observe the pathological changes of the vascular intima and blood clots in different time points after model prepared. The rat model and the rabbit model compare observed and compared with sham operation group do. Results: After preparing model using simple suspension, not only did rat model and rabbit model animal inferior vena venous formed stable thrombus, but also the formation of thrombosis animal survival rate was relatively high, in line with the human vein thrombosis in acute stage of pathologieal evolution. The rabbit model is more simple than rat model. Conclusion: Simple suspension can be successful for the production dynamics of drugs on the formation of deep venous thrombosis (DVT) mechanism in the rabbit or rat deep vein thrombosis model, rabbit is more suitable to the method of modeling.
