大鲵肉对D-半乳糖诱发小鼠衰老相关指标的影响 被引量:6

摘要 目的:探讨大鲵肉对D-半乳糖衰老模型小鼠的抗衰老作用和可能的作用机制。方法:将KM小鼠72只随机分为空白对照组、衰老模型组、阳性对照(Vit E)组和大鲵肉低、中、高剂量组,每组12只。除空白对照组外,其他各组均以D-半乳糖诱发亚急性衰老小鼠模型方法造模,腹腔注射D-半乳糖水溶液125 mg/(kg·d);空白对照组注射同体积注射用水,持续7周;大鲵低、中、高剂量组小鼠分别自由采食16、32、48 mg/(g·d)大鲵肉质,阳性对照组灌胃100 mg/(kg·d)Vit E水溶液,空白对照组灌胃同体积注射用水,持续8周。8周后观察大鲵肉对衰老模型小鼠行为学及基本体征的影响,并检测小鼠血清中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及端粒酶(TE)活性。结果:与空白对照组相比,衰老模型组小鼠体质量增加明显减少,血清SOD和端粒酶活性降低、MDA含量增加(P<0.01);与模型组相比,大鲵肉各剂量组小鼠体质量增加升高,血清SOD活性升高,MDA含量减少(P<0.05);大鲵肉低、中剂量组小鼠血清端粒酶活性有升高趋势,但差异无统计学意义(P>0.05);大鲵肉高剂量组及VE组小鼠血清端粒酶活性明显升高(P<0.05)。结论:大鲵肉对D-半乳糖诱导衰老模型小鼠有抗衰老作用,其机制可能与提高小鼠血清SOD、端粒酶活性,减少MDA含量有关。 Objective: To explore anti-aging effects of giant salamander (Andrias davidianus)meat on D-galactose induced aging mice models and its potential mechanism. Methods: All 72 KM mice were randomized into blank control group, aging model group, positive control (Vit E) group, low, moderate and high doses groups of giant salamander meat, 12 mice each group. Except blank control group, mice models were prepared by peritoneal injection of D-galactose aqueous solution in the concentrations of 125 rag/ (kg'd); blank control group were given water for injections in the same volume for seven consecutive weeks; the mice in low, moderate and high doses groups of giant salamander meat ate giant salamander meat respectively and freely in the concentrations of 16, 32 and 48 mg/ (g. d), positive control group received intragastric administration ofVit E aqueous solution in the concentrations of 100 mg/ (kg. d), and blank control group water for injections in the same volume for eight consecutive weeks. The influence of giant salamander meat on ethology and basic features of aging mice models were observed in eight weeks, the activity of superoxide dismutase (SOD), the contents of malondialdehyde (MDA) and the activity of telomerase (TE) in the serum of the mice were detected. Results: Compared with blank control group, body weight of the mice in the aging model group decreased notably, the activity of SOD and TE lowered, the contents of MDA increased (P〈0.01); compared with the model group, body weight of the mice in different doses groups of giant salamander meat increased remarkably, the activity of SOD increased and the contents of MDA decreased (P〈0.05); the activity of TE in the mice of low and moderate groups showed the tendency of rising, while the difference had no statistical meaning (P〉0.05); the activity of TE in the mice of high doses group of giant salamander meat and Vit E group rose obviously (P〈0.05).Conclusion: Giant salamander meat show anti-aging effect on D-galactose induced aging mice models, its mechanism might be related to increasing the activity of SOD and TE in the serum of the mice and reducing the contents ofMDA.
