人β-防御素-2在肝细胞癌中的表达及其意义

摘要 目的:探讨人β-防御素-2(HBD-2)在肝细胞癌(HCC)中的表达及其意义。方法:收集2011年8月-2014年10月手术切除的原发性肝癌标本45例,肝癌癌旁组织30例,正常肝组织20例,采用免疫组化法检测各标本中HBD-2蛋白表达情况,采用聚合酶链反应(PCR)、实时聚合酶链反应(Real-Time PCR)检测HBD-2 m RNA表达情况,同时采用原位末端标记法(TUNEL法)测定癌组织的细胞凋亡指数。结果:HBD-2 m RNA及HBD-2蛋白在肝癌组织、癌旁组织和正常肝组织中的表达情况比较差异均有统计学意义(P<0.05)。HBD-2蛋白表达阳性的肝癌组织的细胞凋亡指数高于HBD-2蛋白表达阴性的肝癌组织,比较差异有统计学意义(P<0.01)。结论:HBD-2在肝细胞癌组织中低表达,HBD-2可能通过影响肝癌细胞的凋亡而起作用。 Objective: To investigate the expression and significance of HBD-2 in hepatocellular carcinoma ( HCC ) . Method: 45 specimens of HCC tissues, 30 specimens of para-carcinoma tissues and 20 specimens of normal liver tissues in our hospital from August 2011 to October 2014 were collected.The expression situation of HBD-2 protein in the tissues was evaluated by immunohistochemistry, the expression situation of HBD-2 mRNA in the tissues was evaluated by PCR and Real-Time PCR, the cell apoptosis indexes in the tissues were detected by TUNEL methods.Result: The differences in the mean expression of HBD-2 protein and HBD-2 mRNA of hepatocellular carcinoma tissues, para-carcinoma tissues and normal liver tissues were statistically significant ( P〈0.05 ) .The cell apoptosis indexes in the HCC tissues with positive expression of HBD-2 protein was higher than that in the HCC tissues with negative expression of HBD-2 protein,the difference was statistically signifieant( P〈0.01 ). Conclusion: The expression of HBD-2 in HCC is low, it may play a part through the cell apoptosis of HCC.
