摘要
丝氨酸蛋白酶抑制因子Kazal 1型(serine protease inhibitor Kazal type 1,SPINK1)是蛋白酶抑制剂家族中最主要的成员之一,它可抑制胰腺中胰蛋白酶原的非正常性激活,保护胰腺组织,在胰腺炎的发病过程中可能起着重要的作用。除此之外,越来越多的研究表明SPINK1/Spink3(小鼠同源基因)还具有生长因子、自噬的负调节因子等作用,且其基因突变可能是胰腺炎发病的重要危险因素之一,与此同时还发现SPINK1/Spink3可能在肿瘤的发生、发展及预后中起着重要的作用。但有关SPINK1/Spink3发挥其作用的信号传导通路目前尚不清楚。本文就SPINK1/Spink3在以上方面的最新研究进展进行综述。
Serine protease inhibitor Kazal type 1 ( SPINK1 ) is one of the main members of serine protease inhibitor fami- ly. SPINK1 can inhibit abnormal trypsinogen activation, so as to protect pancreatic tissue against the damage. It is believed that SPINK1 possibly plays an important role in the pathogenesis of pancreatitis. Moreover, recent research showed that SPINK1/Spink3 ( a mouse homolog gene of human SPINK1 ) act as the growth factor as well as negative regulator of autophagy. The mutations of SPINK1 gene is probably one of the risk factors for the development of pancreatitis. Besides, SPINK1/Spink3 is likely associated with the occurrence, development and prognosis of cancer. The function - related signaling pathways of SPINKI/Spink3 still remain unclear. In this review, we summarize the roles of SPINK1/Spink3 based on the data obtained from the latest research.
