摘要
目的 :探讨瘦素(leptin)对乳腺癌MCF-7细胞迁移和侵袭能力的影响及其可能的分子作用机制。方法 :用leptin(200 ng/m L)处理乳腺癌MCF-7细胞后,采用划痕愈合实验和Transwell侵袭实验分别检测MCF-7细胞的迁移及侵袭能力,采用实时荧光定量PCR、蛋白质印迹法和ELISA法检测白细胞介素18(interleukin-18,IL-18)的表达水平。用leptin(200 ng/m L)和IL-18结合蛋白(IL-18 binding protein,IL-18bp)(10 ng/m L)同时处理乳腺癌MCF-7细胞后,再采用划痕愈合实验和Transwell侵袭实验分别检测MCF-7细胞迁移及侵袭能力的变化。结果 :Leptin可促进乳腺癌MCF-7细胞的迁移及侵袭(P值均<0.05),上调MCF-7细胞中IL-18 m RNA及蛋白的表达水平(P值均<0.05),促进IL-18分泌(P<0.05)。IL-18bp与细胞分泌的IL-18结合后,leptin促进乳腺癌MCF-7细胞迁移及侵袭的能力被明显抑制(P值均<0.05)。结论 :Leptin可促进乳腺癌MCF-7细胞的迁移及侵袭,其机制可能与促进IL-18表达上调相关。
Oblective: To investigate the effects of leptin on migration and invasion of human breast cancer MCF-7 cells, and to explore its possible molecular mechanism.Methods: After treatment with 200 ng/mL leptin, the migration and invasion abilities of MCF-7 cells were detected by scratch wound healing assay and Transwell invasion test, respectively. The expression level of interleukin-18 (IL-18) in MCF-7 cells after leptin intervention was detected by real-time fluorescent quantitative-PCR,Western blotting and enzyme-linked immunosorbent assay (ELISA). Moreover, the migration and invasion abilities of MCF-7 cells after treatment with combination of 200 ng/mL leptin and 10 ng/mL IL-18 binding protein (IL-18bp) were detected by scratch wound healing assay and Transwell invasion test.Results: Leptin promoted the migration and invasion of MCF-7 cells (both P 〈 0.05), up- regulated the expression levels of IL-18 mRNA and protein (both P 〈 0.05), and promoted the secretion level of IL-18 in MCF-7 cells (P 〈 0.05), However, IL-18bp could inhibit the effects of leptin on migration and invasion of MCF-7 cells (both P 〈 0.05).Conclusion: Leptin promotes the migration and invasion of MCF-7 cells, and its mechanism may be related to the up-regulation of IL-18 expression.
