聚酰胺-胺-Fe_3O_4复合纳米粒子的制备及细胞毒性研究 被引量:1

摘要 采用共沉淀法制得粒径为10~20 nm的超顺磁性四氧化三铁纳米粒子(superparamagnetic iron oxide nanoparticles,IONP),以树枝状高分子聚酰胺-胺(PAMAM)对SIONP包覆修饰得到核壳结构的复合粒子(PAMAMIONP)。通过马尔文粒度仪、扫描电镜、能谱仪、红外光谱仪对材料形貌和结构进行表征,并将含不同类型和浓度的纳米粒子培养液与Hep G2细胞共培养、MTT法来评价粒子细胞毒性。研究结果表明,IONP外部包覆PAMAM之后,粒子在水溶液中的分散性明显增强、毒性显著降低,且氨基封端的复合粒子(PAMAM-IONP)细胞毒性小于酯键封端的复合粒子(MA-PAMAM-IONP)。 We prepared polyamidoamine-modified superparamagnetic iron oxide nanoparticles with amino group and ester groups. The modified nanoparticles were subjected to detailed characterization using Malvern laser particle size analyzer, SEM, energy disperse spectroscopy ( EDS), FTIR. Cytotoxicity studies conducted on HepG2 cells using methyl tetrazolium(MTT) assays revealed that cell viability of different nanopartieles and concentration. These results demonstrate that biocompatible and water dispersion properties of PAMAM-grafted nanoparticles are better than control group.
