摘要
环介导等温扩增技术(Loop-mediated isothermal amplification,LAMP)是一种新型体外等温扩增特异核酸片段的技术,该技术具有高效、快速、特异、低廉、操作简单和结果可视化判定等优点,在基层现场检测中得到广泛应用。传统的LAMP技术已成功应用于病原微生物的快速检测、食品卫生检测、肿瘤检测、原位扩增和胚胎性别鉴定等方面。随着LAMP技术在样本处理、引物设计、特异性产物可视化检测方面日臻完善,改良后的LAMP技术将向多个方向发展,逐步应用于病原微生物活性检测、遗传病和传染病早期诊断等方面。文章就近年来改良LAMP技术原理、特点及在病原检测中的应用进行综述。
Loop - mediated isothermal amplification(LAMP) is a new thermostatic amplify technology of DNA fragements in vitro. It was applied widely in base for it' s advantages, such as high - efficiency, celerity, specificity, lower price, simplicity, visualization and so on. Traditional LAMP technology had been successfully applied to many fileds, like rapid detection of pathogenic microorganisms, food hygiene or tumor, situ amplification, identification of fetus and other sides. LAMP technology refined the samples, primers and visual detection of products, then applied and developed in various 1 fields, especially in active detection of pathogenic microorganisms and the diagnosis of diseases. Finally, the article summarized the theories, characteristics and applications of the improved LAMP recently.
