摘要
研究木犀草素对大鼠心室肌细胞L型钙电流的影响。分离单个大鼠心室肌细胞,应用全细胞膜片钳技术测定钙电流。木犀草素能够剂量依赖性地降低缺氧心肌细胞L型钙电流:木犀草素小剂量组、中剂量组和大剂量组分别使Ica.L的峰值电流由给药前的(921±51)p A减小到给药后的(583.3±54)p A(P<0.05)、(491.2±47)p A(P<0.05)和(276.3±43)p A(P<0.05),降低Ca2+电流幅值的百分率分别为36.7%、46.7%和70%,而在药物被冲洗5 min后,L型钙电流几乎可恢复到正常水平。木犀草素能够剂量依赖性降低缺氧心室肌细胞的L型钙电流。
To explore the effect of luteolin on L-type calcium current in rat ventricular muscle cells. The single rat ventricular cell was seperated,and the technique of whole-cell patch clamp was apply to detect the calcium current. The results showed that luteolin dose dependently inhibited the L-type calcium current in hypoxia treated ventricular myocytes of the rats:in low dosage group,middle dosage group and high dosage group, the peak of Ica.L respectively reduced from (921±51) pA before treatment to (583.3±54) pA(P〈0.05),(491.2±47) pA(P〈0.05) and (276.3±43) pA (P〈0.05) after treatment, the value of electric current Ca2+that decrease in percentage was 36.7%,46.7%and 70%, respectively, and the electric current Ca2+could almost return to the normal level after luteolin was washout. Luteolin could dose-related inhibit the L-type calcium current in rat hypoxia treated ventricular myocytes.
