高压热水-酶法分段提取香菇多糖的研究 被引量:4

EXTRACTION OF LENTINAN BY HIGH PRESSURE-HOT WATER-ENZYMES METHOD

摘要 首先采用高压热水对香菇样品进行浸提,然后再分别采用纤维素酶、果胶酶、菠萝蛋白酶对香菇样本进行酶解,提取香菇多糖,经单因素试验及正交试验获得各种酶的最佳工艺;然后在获得的各种酶的最佳工艺条件下依次分段用高压热水、纤维素酶、果胶酶及菠萝蛋白酶提取香菇多糖.结果表明:高压热水→纤维素酶→果胶酶→菠萝蛋白酶分段提取香菇多糖的提取率高达5.27%,比单独采用高压热水法提高了101.15%. The lentinan has many functions such as inhibiting tumor, lowering blood lipid, antioxidation, lowering blood sugar, enhancing human immunity and so on. The present study was to investigate the optimal extraction process of lentinan by high pressure-hot water-enzymes method. The lentinan was extracted by high pressure hot water firstly,following by eellulose,pectinase and bromelain respectively,and the optimized process was determined by single factor and orthogonal experiments. The results showed that the optimum extraction condition of high pressure-hot water cooperation with cellulose was as follows: the temperature of 40 ℃ ,pH of 5.5,cellulose concentration of 2%, and the extraction time of 0.5 h, while the extraction rate was up to 4.16%; High pressure-hot water and pectinase was: temperature of 40 ℃, pH of 4.5 ,pectinase concentration of 1.0%, time of 1.5 h,while the extraction of rate was up to 3.39% ;high pressure-hot water and bromelain was : the temperature of 60 ℃ ,pH of 3.5 ,bromelain concentration of 1.0% ,time of 3.5 h,while the extraction rate was up to 2.65%. It was concluded that the extraction rate of lentinan could achieve to 5.27% using the high pressure-hot water, cellulase,pectinase,bromelain fractional-step method and the extraction rate increased by 101.15% compared to high pressure-hot water extraction only. High pressure-hot water--enzymatic method was the optimized method for the lentinan extraction, which was the basis for the comprehensive application of lentinan.
