循环纤维细胞在肾功能衰竭大鼠早期肾脏纤维化中的意义 被引量:1

Significance of circulating fibrocytes in early-stage renal fibrosis of renal failure rats

摘要 目的 通过外周血纤维细胞计数,观察外周血纤维细胞数量与肾脏纤维化程度之间的关系,探讨外周血纤维细胞作为肾纤维化早期生物学标志物及预后指标的可行性.方法 以CD45和Ⅰ型胶原作为循环纤维细胞的表面标记,使用流式细胞仪对5/6肾切除肾衰大鼠外周血纤维细胞计数,通过免疫组化对循环纤维细胞在肾衰大鼠肾脏中的浸润进行鉴定和定位,结合肾功能生化指标,阐述循环纤维细胞计数在肾纤维化中的意义.结果 肾衰大鼠模型组在造模后的第4、8、12周,外周血纤维细胞计数显著升高(P<0.01).造模后外周血纤维细胞计数与肾脏病理改变程度呈正相关(r =0.613,P<0.0l),与血清肌酐(Scr)、尿素氮(BUN)呈正相关(r=0.623,0.550,P<0.01).结论 外周血纤维细胞计数在肾纤维化早期与肾脏病理改变一致,可早期预示肾纤维化的发展进程. Objective To investigate the relationship between the number of peripheral blood fibrocytes and the degree of renal fibrosis,and explore the feasibility of peripheral blood fibrocytes as the early biomarker and prognostic indicator of renal fibrosis.Methods CD45 and type Ⅰ collagen were taken as the markers of circulating fibrocytes.The flow cytometer was used to count bibrocytes in rat peripheral blood with 5/6 kidney cut and carry out identification and positioning on the infiltration of circulating fibrocytes in the kidney of rat with renal failure with immunohistochemistry,and set forth thc significance of circulating fibrocyte counting in renal fibrosis combined with biochemical markers of renal function.Results For the model group of rat with renal failure,in the forth,eighth,and twelfth weeks after modeling,the peripheral blood fibrocyte counting was significantly increased (P <0.01).In the second,forth,and twelfth weeks after modeling,the blood cell counting was significantly positively correlated with renal pathology changes (r-0.613,P < 0.01),and also was significantly positively correlated with serum creatinine (Scr),and blood urea nitrogen (BUN) (r =0.623,0.550,P < 0.01).Conclusions The peripheral blood fibrocyte counting was positively correlated with renal pathology changes in early renal fibrosis and might early indicate the developmental process of renal fibrosis.
