摘要
目的研究TGFβ1与Smad2/3在乳腺癌组织中的表达,揭示其在乳腺癌中的信号转导通路。方法应用免疫组化SABC法检测62例乳腺癌组织中TGFβ1与Smad2/3的表达情况,利用SPSS19.0统计学软件分析TGFβ1与Smad2/3的差异性及相关性。结果 TGFβ1与Smad2/3在乳腺癌组织中呈高表达,阳性表达率依次为69.35%(43/62)和40.32%(25/62)。TGFβ1与Smad2/3在乳腺癌组织中的表达与年龄无关,TGFβ1与Smad2/3在组织学分级、病理学分类、TNM分期方面差异均有统计学意义(P<0.05)。TGFβ1与Smad2/3在乳腺癌组织中的表达呈正相关(r=0.261,P<0.05)。结论乳腺癌的发生可能与TGFβ1及Smad2/3信号转导途径发生异常,致使下游信号Smad2/3突变,增加瘤细胞与细胞外基质的相互作用、抑制免疫系统、增加血管形成等机制促进乳腺癌的浸润和转移有关。
Objective To study the expression of TGFβ1 and Smad2/3 in breast cancer tissues, and to demonstrate its signal transduction pathway in breast cancer. Methods Immunohistochemical SABC method was applied to test the ex- pression of TGFβ1 and Smad2/3 in 62 cases of breast cancer tissues. The statistically software of SPSS19.0 was applied to analyze the difference and correlation between TGFβ1 and Smad2/3. Results TGFβ1 and Smad2/3 were highly expressed in breast cancer tissues, and the positive expression rate was 69.35% (43/62) and 40.32% (25/62) respectively. The ex- pression of TGFβ1 and Smad2/3in breast cancer tissues was not correlated with the age, and the differences of TGFβ1and Smad2/3 in histological grade, pathological classification and TNM stage were all statistically significant (P〈0.05) The expression of TGFβ1 and the expression of Smad2/3 in breast cancer tissues were positively correlated (r=0.261, P〈0.05) Conclusion The possible reason of the occurrence of breast cancer is that the abnormal signal transduction pathway of TGFβ1 and Smad2/3, induces the mutation of downstream signal Smad2/3, and mechanisms such as strengthening interac- tions between tumor cells and extracellular matrix, inhibition of immunological system and improving angiopoiesis are used to promote the infiltration and metastasis of breast cancer
