新疆维吾尔族PS基因多态性及血浆可溶性P选择素水平与心房颤动合并血栓栓塞的关联研究 被引量:2

摘要 目的探讨新疆地区维吾尔族P选择素(PS)基因多态性及血浆可溶性P选择素(sPs)水平与心房颤动(房颤)合并血栓栓塞的相关性。方法选取新疆医科大学第一附属医院综合心脏内科收住的维吾尔族非瓣膜性房颤患者302例,按是否合并血栓栓塞分为病例组(房颤合并血栓栓塞患者,103例)和对照组(房颤无血栓栓塞患者,199例),采用酶联免疫吸附试验(ELISA)方法测定其血浆中可溶性P选择素(sPs)浓度,并采用聚合酶链反应(PCR)和限制性片断长度多态性,分析PS基因在2123C/G位点、Thr715Pro位点的多态性与房颤血栓栓塞形成的相关性。结果病例组患者血浆sPs的平均浓度为(42.4±8.2)μg/mL,高于对照组患者血浆sPs平均浓度(36.3±8.24)μg/mL,差异有统计学意义(P=0.001)。PS基因2123C/G位点存在多态性,病例组和对照组基因型和等位基因分布差异有统计学意义(χ2=13.275,P=0.001;χ2=11.693,P=0.001)。血浆sPs水平与2123C/G基因多态性有明显相关性,GG基因型者的血浆sPs水平显著高于GC+CC基因型(P<0.05)。未检测出Thr715Pro基因多态性。结论血浆sPs水平和PS基因多态性与新疆维吾尔族心房颤动合并血栓栓塞形成存在一定的关联。 Objective To investigate the association of PS gene polymorphisrn and soluble P-selectin levels in atrial fibrillation thrornboernbolisrn in Xinjiang Uygur. Methods 302 Uighur patients who were hospi- talized patients with nonvalvular atrial fibrillation were selected as subjects in xinjiang medical university first affiliated hospital heart center , and were divided into the case group (merger thromboernbolisrn group, 103 cases) and the control group (no thrornnboembolisrn group, 199 cases) according to whether or not the merger thromboembolisrn. Using ELISA method determination of plasma level of sPs. The fre- quency distributions of SNP sP-selectin gene promoter (- 2123 C/G)and SNP in exon region (Thr715Pro) were investigated by polymerase chain reaction (PCR)-restriction fragment length polymor- phism and direct DNA sequence analysis. Results The average plasma concentration of 103 cases in thecase group sPs (42.4±8.2) μg/mL, higher than sPs 199 patients in the control group (36.3±8.24) μg/mL, the difference was statistically significant (P 〈0.001). PS gene 2123 C/G polymorphism loci exist, the genotype and allele distribution difference among the case group and the control group were statistically significant (χ2=13.275, P =0.001; χ2 =11.693, P =0.001). sPs serum level and 2123 C/G gene polymor- phism have obvious correlation, GG genotype of sPs serum level was significantly higher than that of GC+ CC genotype (P 〈0.05). No ThrT15Pro polymorphism was detectable. Conclusion The sP-selectin gene polymorphisms might have association with serum sP-selectin levels or thromboembolic events.
