坤泰胶囊对去卵巢大鼠骨质疏松防治效果的实验研究 被引量:7

摘要 目的研究坤泰胶囊对去卵巢大鼠骨质疏松的防治效果,旨在为科学用药提供依据。方法随机取60只4月龄雌性大鼠,10只大鼠作为假手术组,50只大鼠进行去卵巢造模。术后4周将建模成功的50只雌性SD大鼠随机分成模型组、坤泰胶囊低剂量组、坤泰胶囊中剂量组、坤泰胶囊高剂量组及仙灵骨葆胶囊组,每组10只。假手术组及模型组灌胃等容积的蒸馏水,坤泰胶囊低、中、高剂量组以及仙灵骨葆胶囊组分别灌胃坤泰胶囊1.0 g/kg、2.0 g/kg、4.0 g/kg及仙灵骨葆胶囊2.0 g/kg,1次/d,连续12周。观察各组大鼠体质量、骨密度以及血清骨碱性磷酸酶、抗酒石酸酸性磷酸酶5b水平。结果坤泰胶囊中、高剂量组的体质量及血清骨碱性磷酸酶、抗酒石酸酸性磷酸酶5b水平均明显低于模型组(P均<0.05),骨密度值显著高于模型组(P均<0.05)。各指标与仙灵骨葆胶囊组相当。结论中、高剂量坤泰胶囊可以有效增加去卵巢骨质疏松大鼠骨密度值,降低血清骨碱性磷酸酶、抗酒石酸酸性磷酸酶5b水平,是治疗绝经后骨质疏松症的有效药物。 Objective It is to study the effect of prevention and treatment of Kuntai Capsule on osteoporosis in ovariectomized rats,in order to provide a basis for scientific use. Methods Ten 4 months old female rats were randomly selected as sham operation group,another 50 rats were selected to establish ovariectomized models. In 4 weeks after the operation,the successful models of 50 rats were divided into model group,low,medium and high dose of Kuntai capsule groups and Xianlinggubao capsule group,each group had 10 rats. The sham operation group and model group was given the same volume of distilled water by intragastric administration,low,medium and high dose of Kuntai capsule groups and Xianlinggubao capsule group was respectively treated with Kuntai capsule 1. 0 g / kg,2. 0 g / kg,4. 0 g / kg and Xianlinggubao capsule 2. 0 g / kg by intragastric administration once per day and for 12 weeks. Bone density,body mass,and the levels of serum biochemical markers of bone alkaline phosphatase,tartrate resistant acid phosphatase 5b of the rats in every group were observed. Results Body mass and the levels of serum bone alkaline phosphatase,tartrate resistant acid phosphatase 5b in Xianlinggubao capsule group and medium and high dose of Kuntai capsule groups were significantly lower,while bone density was higher than those in the model group( P〈0. 05),and the changes of these indexes were similar in Xianlinggubao capsule group and medium and high dose of Kuntai capsule groups. Conclusion Medium and high dose of Kuntai capsule can effectively increase bone density,decrease the levels of serum bone alkaline phosphatase,tartrate resistant acid phosphatase 5b,so it is the effective drug for the treatment of postmenopausal osteoporosis.
