明日叶查尔酮对急性辐射损伤小鼠氧化损伤的作用研究 被引量:3

摘要 目的:研究明日叶查尔酮对小鼠急性辐射氧化损伤及细胞增长与凋亡的防护作用。方法:将50只昆明种小鼠随机分五组,每组10只。低、中、高剂量组每天灌胃给予明日叶查尔酮4、20、40 mg/kg,辐射损伤组与空白对照组给予生理盐水,连续5 d,第6天给予一次性X射线全身照射4 Gy,空白对照组不照射。照射后按前述方法继续灌胃5天处死动物,检测各组小鼠肝脏总抗氧化能力(T-AOC)、脂质过氧化物(MDA)、超氧化物歧化酶(SOD)及肝细胞Bcl-2蛋白表达水平。结果:与正常对照组比较,辐射损伤组T-AOC、SOD及Bcl-2降低,MDA含量增高,高剂量组较辐射损伤组T-AOC、SOD及Bcl-2升高,MDA含量降低,上述各组的差异均有显著性意义(p<0.05)。结论:明日叶查尔酮对小鼠X射线所致的氧化损伤有一定防护作用。 objective:to study the Ashitaba Chalcone in mice with acute radiation oxidative damage and protection of cell growth and apoptosis. Methods :50 healthy kunming mice were randomly divided into 5 groups ,which include 10 rats in each group. AC high,middle and low dose group were respectively fed through mouth with ashitaba chalcone in concentration of 40, 20, 4rag/( kg· bw)/d , while normal con- trol group and radiation control group were fed through mouth with distilled water, oral administration of 0.2ml/20g body weight. After continuous intragastric administration for 5 days, all mice were exposed to 3 Gray X-radiation one time except for nomal control group and then all the mice continued to be fed for 5 days according to the above methods. At the end of the experiment, the mice were killed. The parameters that need to be measured include all groups of mice liver total antioxidant capacity (T-AOC) and lipid peroxide (MDA), superoxide dismutase (SOD) and liver cell the Bcl- 2 protein expression level. Re- suits:Compared with normal control group, contents of T-AOC,SOD and Bcl-2 decreased and MDA in- creased in radiation control group. While compared with radiation control group, contents of T-AOC, SOD and Bcl-2 increased and MDA decreased in AC high dose group. These difference were statistically signifi- cant(p 〈 0.05 ). Conclusion : Ashitaba Chalcone (AC) have some protective effect on X-ray radiation inju- ry in mice.
