摘要
组蛋白赖氨酸甲基转移酶SET8属于SET基因家族成员,是目前发现的唯一可以特异性催化H4赖氨酸20单甲基化(H4K20me1)的赖氨酸甲基转移酶(KMTs);此外,SET8还可甲基化p53、TWIST、Wnt及ERα等非组蛋白,并通过调节转录过程影响相应基因的表达,进而参与调控细胞周期、染色质固缩和DNA的复制。有研究提示,SET8的单核苷酸多态性与多种肿瘤的发生发展有潜在的相关性。本文就SET8对组蛋白和非组蛋白的修饰、micro RNA对SET8的调节及SET8与肿瘤相关性的研究进展进行了综述,旨在为揭示肿瘤的发病机制和筛选治疗靶点提供帮助。
SET8 is a member of SET gene family. SET8 is the only histone methyltransferase (KMT) that can uniquely catalyze histone monomethylation of H4 lysine 20 (H4K20me1). Furthermore, SET8 can methylate other non-histone proteins, such as p53, TWIST, Wnt, and ERα. SET8 can affect the expression of the corresponding genes through gene transcription regulation. SET8 subse-quently contributes to the regulation of gene transcription, cell cycle, chromatin condensation, and DNA replication. Population studies suggested that a single nucleotide polymorphism on SET8 gene is potentially associated with the development of various tumors. In this review, we focus on research progress on SET8 function in histone and non-histone modifications, microRNA regulatory role on SET8, and SET8's correlation with tumor. We aim to reveal cancer pathogenesis and contribute to the screening of therapeutic targets.
