摘要
目的预测RhD抗体核酸适配体序列库中优势适配体,为实验室验证做准备。方法对以RhD抗体为目标靶分子而筛选出的适配体序列库做克隆测序;使用ClustalⅩ软件对阳性克隆子序列做同源性分析;使用DNA folding form在线分析工具对阳性克隆子碱基序列做最小自由能及二级结构分析。结果 RhD抗体28个阳性克隆子碱基序列按照基序的不同可分为3个家族,家族1和2中的适配体又可分为不同的亚家族;可形成由2-4个多分支环组成的二级结构,最小自由能在(-7.47--5.09)kcal/mol。结论 RhD家族1中的亚家族3和4适配体二级结构较为稳定,最小自由能较低,是RhD抗体适配体库中的优势适配体。
Objective To predict the advantage of aptamers in RhD antibody aptamer-library in order to prepare for lab test. Methods RhD antibody aptamer base sequence was analyzed by cloning sequencing. The homology sequence,minimum free energy and secondary structure of aptamers were analyzed via Clustal X and DNA folding form software. Results28 positive clones could divide into three families according to different motifs. Aptamers in family 1 and 2 could divide into different subfamilies. All of them could form 2-4 branched loop structures and minimum free energy was between-7. 47-5. 09 kcal / mol. Conclusion Aptamers in family 1 subfamily 3 and 4 are aptamers with stable structure and lower minimum free energy in RhD antibody aptamer-library
