储存血液中CD4^+ CD25^+调节性T细胞表达水平的研究

摘要 目的探讨储存血液中调节性T细胞表达的变化及意义。方法采用流式细胞术检测5名健康献血者新鲜血液及储存3周后血液中CD4+CD25+调节性T细胞及CD4+CD25+Foxp3+的变化。结果新鲜血液与储存3周后血液相比CD4+CD25+调节性T细胞的值与CD4+CD25+Foxp3+的值均降低(P<0.01)。新鲜血液与储存3周后血液CD4+CD25+Foxp3+表达成线性相关。结论储存血液调节性T细胞及其基因Foxp3表达的明显降低可能为临床相关疾病输血治疗及疗效的评估提供了新的思路。 Objective To investigate expression of regulatory T cell CD4 ^+ CD25^+ in stored blood. Methods The expression of CD4 ^+ CD25^+ , and CD4 ^+ CD25^+ Foxp3^+ T cells of the blood samples, collected from 5 healthy blood donors, were tested as soon as collected and 3-week after storage via flow cytometry. Results The value of CD4 ^+ CD25^+ regulatory T ceils, as well as CD4 ^+ CD25^+ Foxp3^+ in fresh blood decreased, when compared to the blood after 3-week storage(P 〈 0. 001 ), with statistical significance. The CD4 ^+ CD25^+ Foxp3^+ expression in fresh blood was linearly collected with the expression in blood after 3-week storage. Conclusion The significant decrease of the expression of regulatory T ceils and Foxp3 gene in stored blood could provide some new ideas to the clinical treatment and assessment of curative effect.
