外源正常淋巴液对大鼠MI/RI损伤影响

Effect of exogenous normal lymph on myocardial ischemia-reperfusion injury in rats

摘要 目的:研究外源正常淋巴液对心肌缺血再灌注损伤(MI/RI)大鼠心电图及病理改变和炎性因子的影响,探讨正常淋巴液在MI/RI中的治疗效应。方法:45只雄性SD大鼠随机分为假手术组、血浆组、淋巴液组。除假手术组外,均采用冠状动脉左前降支根部穿线结扎复制大鼠MI/RI模型。淋巴液组输入淋巴液,假手术组及血浆组用血浆代替。监测缺血期与再灌注期心电图的改变,24h后取心肌组织行病理切片,并测定血浆肿瘤坏死因子(TNF-α)、过氧化物歧化酶(SOD)、白介素6(IL-6)水平。结果:与血浆组相比,淋巴液组心梗后30min ST段抬高度低,心率快,再灌注30min后ST段低,心率快(P<0.05)。与假手术组相比,再灌注24h后淋巴液组及血浆组大鼠血浆中TNF-α、SOD、IL-6均升高,与血浆组相比,淋巴液组TNF-α、IL-6降低,SOD升高(P<0.05)。结论:正常淋巴液可能通过减少炎性反应和中性粒细胞浸润,降低TNF-α、IL-6水平,维持SOD活力这一途径减轻炎症、氧化应激介导的心肌缺血再灌注损伤,但是可能会加重钙反常。 Objective.. To investigate the treatment effect of normal lymph on myocardial ischemia reperfusion injury through studying the effect of exogenous normal lymph on the electrocardiogram (ECG), pathological and inflammatory factor of rats with myocardial ischemia-reperfusion injury (MI/RI). Methods. 45 male SD rats were randomized into the control group, the plasma group, and the lymph group. In addition to the control group, coronary artery ligation of left anterior de scending branch root with thread were used in the other 2 groups to copy MI/RI model in rats. The lymph group was injected with lymph, and plasma was used in the control group and the plasma group instead. The changes of electrocardiogram (ECG) in the stages of ischemia and reperfusion were monitored, then the myocardial tissues were taken after 24 h for pathological slices and the tumor necrosis factor (TNF-α), superoxide dismutase (SOD) and interleukin 6 (IL-6) levels in the plasma were detected. Results: Compared with the plasma group, lymph group show a faster heart rate and low ST segment lifting height 30 min after myocardial infarction, and also low ST segment and faster heart rate 30 min after reperfusion (P(0.05). Compared with the control group, TNF a, SOD and IL-6 levels in the lymph group and the plasma group 24 h after the reperfusion in creased; and compared with the plasma group, TNF alpha, IL-6 levels reduced; while SOD in creased in the lymph group (P〈0.05). Conclusion. By reducing inflammatory reactions and neu trophil infiltration, normal lymph may reduce the levels of TNF-α, IL-6, and maintain the activity of SOD to reduce ischemia-reperfusion injury which was mediated by inflammation and oxidative stress, but it may increase the calcium paradox.
