摘要
目的:研究肝癌患者血清中CD44v6、HMGB1以及GP73含量及其与肿瘤组织中JAK-STAT通路的相关性。方法:选择在本院确诊为原发性肝癌的60例患者作为观察组,将同期在本院体检明确身体健康的60例健康者作为对照组,采集血清后检测CD44v6、HMGB1、GP73含量,收集肝癌和癌旁组织并检测JAK-STAT信号分子及下游调节基因的mRNA含量。结果:与对照组比较,观察组血清中CD44v6、HMGB1以及GP73的含量较高,差异有统计学意义(P<0.05);肝癌组织中JAK1、JAK2、STAT1、STAT3、STAT5的表达量高于癌旁组织,差异有统计学意义(P<0.05);肝癌组织中TCF21、Pim1、VEGF、HIF-1α、ICAM-1、VCAM-1的表达量高于癌旁组织,差异有统计学意义(P<0.05);血清CD44v6、HMGB1以及GP73含量与JAK1、JAK2、STAT1、STAT3、STAT5的表达量呈正相关。结论:肝癌患者血清中肿瘤标志分子血清中CD44v6、HMGB1以及GP73含量异常升高且与肝癌组织中JAK-STAT通路的激活密切相关。
Objective:To study the serum CD44v6,HMGB1,GP73 contents of liver cancer patients and its correlation with JAK-STAT pathway in tumor tissue.Methods:60cases of patients diagnosed as primary liver cancer in our hospital from2013/6-2014/8were enrolled as observation group,60 cases of healthy people judged by health examination in our hospital during the same period were enrolled as control group.Then serum was collected and CD44v6,HMGB1,GP73 contents were assayed,hepatocellular carcinoma tissue and paracancerous tissues were collected and JAK-STAT signal molecules and downstream regulatory gene were assayed.Results:(1)Serum indexes:compared with serum hepatocellular carcinoma marker molecules,CD44v6,HMGB1,GP73 contents in serum of observation group were lower;(2)JAK-STAT pathway:JAK1,JAK2,STAT1,STAT3,STAT5 expression in hepatocellular carcinoma tissue were higher than those in paracancerous tissues;(3)Signaling pathway regulates gene:TCF21,Pim1,VEGF,HIF-1α,ICAM-1,VCAM-1expression in hepatocellular carcinoma tissue were higher than those in paracancerous tissues;(4)Serum CD44v6,HMGB1 and GP73contents were positively correlated with JAK1,JAK2,STAT1,STAT3,STAT5 expression.Conclusion:Serum tumor marker molecules CD44v6,HMGB1 and GP73contents of liver cancer patients abnormally increase and have correlation with the activation of JAK-STAT pathway in hepatocellular carcinoma tissue.
