摘要
目的研究补骨脂中9种黄酮类化合物构效关系及对成骨细胞的增殖、分化活性。方法采用从绝经后妇女股骨松质骨分离得到的成骨细胞,利用噻唑蓝法检测黄酮类化合物对成骨细胞增殖的影响,同时测量成骨细胞碱性磷酸酶(ALP)活性来评价黄酮类化合物对人成骨细胞分化的影响。结果 9种黄酮化合物中,补骨脂二氢黄酮药物组对成骨细胞增殖、分化的活性最强,通过对9中化合物的EC50值和各药物组成骨细胞酶活性比较,发现二氢黄酮母核、7位羟基、6位异戊二烯基的存在对成骨细胞的增殖、分化具有显著的作用。结论补骨脂中黄酮类化合物对成骨细胞的增殖、分化作用与其结构有一定的相关性。
OBJECTIVE To investigate the effects of nine flavonoids in Buguzhi on the proliferation, differentiation of on Postm- enopausal women's osteoblasts and their structure-activity relationship analysis. METHODS Osteogenie cells were obtained from the femur in postmenopausal women. The effects of flavonoids on the proliferation, differentiation and osteoblasts were determined by MTY method, measuring the activity of alkaline phosphatase. RESULTS Among the nine flavonoids, bavachin exhibited the highest proliferation, differentiation activity. The final result showed that their proliferation, differentiation activity may be related to the parent structure, the position of hydroxyl group and prenyl group. The results demonstrate that flavonone parent, 7-OH, 6-prenyl may improve the proliferation, differentiation effect. CONCLUSION The results demonstrated that the effects of nine flavonoids in Buguzhi on proliferation, differentiation activity had connection with their structures.
