摘要
目的观察缺血/缺氧(I-H)预处理在子宫内膜异位症中的作用及机制。方法诊断子宫肌瘤需行子宫切除手术患者20例,月经周期正常,病理证实子宫内膜处于分泌早期。术中分离双侧子宫动脉,采用无损伤血管夹钳夹/开放子宫动脉各5min,重复3次。I-H预处理前(对照组)、后(实验组)刮取前壁子宫内膜,种植于鸡胚尿囊膜(CAM),72h后观察种植内膜成活率。免疫组化检测内膜组织中基质金属蛋白酶1(MMP-1)的表达。结果成功构建了鸡胚模型。实验组种植灶中MMP-1的含量高于对照组(P<0.05)。结论 I-H预处理上调子宫内膜MMP-1的表达,提高CAM模型中种植成活率。
Objective To study the role and mechanism of ischemic‐hypoxic (I‐H ) preconditioning improving planting survival rate of human endometrium in chick chorioallantoic membrane(CAM) model .Methods The human endometrium was taken from 20 women with uterine fibroids during hysterectomy ,who were confirmed to have normal menstrual cycle and the endometrium was confirmed pathologically to be in the early secretory phase .I‐H preconditioning of endometrium was performed by clamping and opening bilateral uterine arteries for three times .Every procedure of clamping or opening lasted for 5 minutes .The endometrium was taken before(group A) and after I‐H preconditioning(group B) and planted into CAM .The planting survival rate of human endometrium in CAM was observed at 72 hours after planting .The expression of matrix metalloproteinase‐1(MMP‐1) in the cultivation of CAM was detected by immunohistochemical assay . Results The CAM model was constructed successfully .The expression of MMP‐1 in group B was much higher than that in group A ( P〈0 .05 ) .Conclusion I‐H preconditioning of endometrium upregulates MMP‐1 expression and improves planting survival rate of human endometrium in CAM model .
