摘要
[目的]用B交配型位点基因对杏鲍菇不同极性菌株进行快速鉴定。[方法]根据杏鲍菇B交配型位点基因的信息素PEphb(3.3.1~3.3.4)和信息素受体PEPHSTE(3.3.1~3.3.4)基因设计8对特异性引物,对原生质体单核化得到的2个不同极性的杏鲍菇样本进行PCR扩增。[结果]4个信息素基因和3个信息素受体基因仅能在A2B2极性中存在,而在A1B1极性中不存在。[结论]用B交配型位点基因能对杏鲍菇不同极性进行鉴定,且这7个基因可以作为杏鲍菇原生质体单核化极性快速鉴定的SCAR分子标记。
[ Objective]The aim of this study was to identified different polarity of P. eryngii using B mating locus genes. [ Methods ] The pheromone genes PEphb ( 3.3.1 - 3.3.4) and pheromone receptors genes PEPHSTE ( 3.3.1 - 3.3.4 ) from B mating- type locus of P. eryngii were used to designed PCR primers, PCR amplification using different polarmonokaryons strains as templets. [ Results ] Four pheromone genes PEphb (3.3.1,3.3.2,3.3.3 and 3.3.4) and three pheromone receptors PEPHSTE( 3.3.1,3.3.2 and 3.3.4) only exist in A2B2 polarity monokaryons strains. [ Conclusion] Identified different polarity of P. eryngii using B mating - type locus and these seven genes can be used as SCAR markers of P. eryngii protoplast monokaryons.
