葡萄糖调节蛋白-78和磷酸化真核翻译启动因子2α在膀胱尿路上皮癌中的表达及临床意义

摘要 目的分析葡萄糖调节蛋白-78(GRP78)及磷酸化真核翻译启动因子2α(P-e IF2α)在正常膀胱黏膜组织和膀胱泌尿上皮癌组织中的表达。方法采用Western blot法检测16对膀胱癌和正常膀胱活检新鲜组织的GRP78和P-e IF2α蛋白的表达水平。采用免疫组织化学方法检测67例膀胱泌尿上皮癌组织中GRP78蛋白的表达,并分析其蛋白表达水平与临床病理特征之间的相关性。结果 GRP78蛋白在膀胱泌尿上皮癌中表达较正常膀胱黏膜组织中上升。免疫组织化学染色结果显示膀胱泌尿上皮癌组织中的GRP78的表达水平与临床分期、浸润深度及淋巴结转移呈正相关(P<0.05)。结论在膀胱泌尿上皮癌中,GRP78较正常组织中表达上调,并且与膀胱泌尿上皮癌的浸润深度及淋巴结转移相关,P-e IF2α在癌组织与正常组织中表达无显著差异。 Objective To analyze the expression and significance of GRP78 and P-eIF2α in bladder urothelial carcinoma and normal mucosa. Methods Western blot was used to detect expression of GRP78 and P-eIF2α levels in 16 couples of cancer tissue and normal mucosa. Immunohisto- chemistry was used to detect expression of GRP78 level in 67 formalin-fixed paraffin-embedded bladder urothelial carcinoma, and correlation between GRO78 level and clinical pathological features was analyzed. Results The results showed that the expression of GRP78 was up-regulated in cancer tissue, and the level of GRP78 expression was correlated with the depth of invasion, positive lymph node metastasis and TNM stage. Conclusion In bladder urothelial carcinoma, level of GRP78 is up- regulated when compared with normal tissue, and is correlated with the depth of invasion and lymphatic metastasis of bladder urothelial carcinoma, but the expression of P-eIF2α shows no significant difference in carcinoma tissue and normal tissue.
