摘要
目的探讨多西他赛联合顺铂对人肺癌细胞株(A549细胞)热休克蛋白70(HSP70)表达的影响。方法体外培养肺癌A549细胞,分别采用多西他赛20μg/m L、顺铂5μg/m L及二者联合用药处理细胞,另设不加药物处理的对照组,MTT法检测A549细胞增殖活力;同时采用Western Blot检测顺铂、多西他赛及二者联合用药分别处理细胞后,HSP70表达量的变化。结果 MTT法显示,多西他赛、顺铂、多西他赛联合顺铂处理A459细胞后,细胞增殖明显受到抑制,细胞活性明显低于对照组(P<0.01);联合组细胞活性显著低于多西他赛组和顺铂组(P<0.01);Western Blot显示,多西他赛组、顺铂组、联合组HSP70表达均降低(P<0.05或P<0.01);联合用药组HSP70表达低于单独用药组,差异均具有统计学意义(P<0.05)。结论多西他赛与顺铂均具有抑制肺癌A549细胞的增殖作用,二者联用具有协同作用,其机制可能与下调HSP70的表达有关。
Objective To explore the influence of docetaxel combined with platinum on the expression of heat shock protein 70 (HSP70) in human lung cancer cell line (A549 cells). Methods Lung cancer A549 cells were in - vitro cultured and treated with docetaxel 20μg/mL (docetaxel group), platinum 5 μg/mL (platinum group) and docetaxel combined with platinum (combination group), respectively, and control group without drug treatment was established. Methyl thiazolyl tetrazolium (MTT) was used to detect the activity of A549 cell proliferation and Western Blot to detect the changes of HSP70 expression after treated with docetaxel, platinum and docetaxel com- bined with platinum, respectively. Results MTT method showed that after treatment, cell proliferation was evidently inhibited in all groups and cell activity was markedly lower than control group (P 〈 0.01 ), and combination group was prominently lower than docetaxel group and platinum group in cell activity (P 〈 0.01). Western Blot results indicated that HSP70 expression decreased obviously in all groups (P〈0.05, P〈0.01), and was notably lower in combination group than in other two groups (P 〈 0.05). Conclusion Both docetaxel and platinum can inhibit the proliferation of lung cancer A549 cells, and their combined application has synergetic action, whose mechanism is in association with the down - regulation of HSP70 expression.
