miR-218靶定SOX5对胶质瘤细胞生长的影响 被引量:2

摘要 目的观察micro RNA-218(miR-218)靶定SOX5对胶质瘤细胞生长的影响。方法首先利用生物信息学方法、荧光素酶报告系统验证SOX5是miR-218直接作用靶点。通过RT-PCR和Western blot方法检测胶质瘤细胞系、胶质瘤组织及其瘤旁正常脑组织中miR-218对SOX5的mRNA和蛋白水平的影响。然后在胶质瘤U87细胞系中,过表达miR-218后利用MTT检测胶质瘤细胞系表型的变化。结果利用生物信息学预测出基因SOX53,UTR含有miR-218的结合位点,荧光素酶报告结果进一步证实miR-218靶定SOX5。RT-PCR结果显示,与正常脑组织相比,胶质瘤组织内miR-218的表达明显降低,而SOX5的mRNA明显增高。MTT结果显示,在U87细胞系中,过表达miR-218后,细胞生长活性下降,沉默SOX5后胶质瘤细胞的增殖活性都明显下降。结论 SOX5是miR-218的直接作用靶点,miR-218有可能通过抑制SOX5基因的表达发挥抑癌基因的作用。
