摘要
含硫生物分子在自然界中普遍存在并参与重要的生物化学反应。半胱氨酸脱硫酶在辅因子磷酸吡哆醛(PLP)作用下催化底物L-半胱氨酸脱硫生成L-丙氨酸及酶过硫化物中间体。半胱氨酸脱硫酶除用于铁硫簇的组装外还参与众多含硫化合物的合成如:t RNA的硫修饰、DNA硫修饰、硫胺素的合成以及钼喋呤辅因子合成等等。本文综述了半胱氨酸脱硫酶的结构及功能的最新研究进展,以期能为后续半胱氨酸脱硫酶的研究提供参考。
Sulfur-containing cofactors are widely distributed in nature and participate in essential biochemi- cal reactions. Cysteine desulfurases utilize a PLP-dependent mechanism to catalyze the conversion of L-cyste- ine to L-alanine and sulfane sulfur via the formation of a protein-bound cysteine persulfide intermediate on a conserved cysteine residue. Cysteine desulfurases participate in many biosynthesis ofthio-cofactors, including Fe-S clusters, thionucleosides, DNA sulfur modification, thiamin, molybdenum cofactor and so on. We review the recent advance of structure and functions of cysteine desulfurases in order to provide the reference for lat- er study of cysteine desulfurases.
