小动物PET/CT显像考察麻黄碱干预棕色脂肪^(18)F-FDG摄取的研究 被引量:1

摘要 目的:采用小动物PET/CT研究不同剂量麻黄碱(Ephedrine,Eph)与温度对棕色脂肪(Brown Adipose Tissue,BAT)摄取18F-氟代脱氧葡萄糖(18F-fluoro-deoxyglucose,18F-FDG)的影响。方法:将昆明小鼠腹腔注射18F-FDG或18F-FDG+Eph(40 mg/kg、80 mg/kg和160 mg/kg),放置于不同实验温度条件下,1 h后采用小动物PET/CT对小鼠进行全身PET/CT扫描,通过Inveon Research Workplace(IRW)软件对小鼠肩胛处BAT摄取18F-FDG的标准化摄取值(Standardised uptake value,SUV)进行分析。结果 :25℃和15℃未使用Eph干预的BAT SUV值分别为1.67±0.49和2.7±0.26;25℃和15℃使用Eph(40 mg/kg)干预BAT SUV值分别为2.20±0.36和5.97±0.21。温度为25℃,Eph分别为40 mg/kg、80 mg/kg和160 mg/kg时BAT SUV值分别为2.20±0.36、3.33±0.31和6.73±0.50。结论 :低温可以刺激BAT的能量代谢增高,同时随着Eph的剂量增高BAT的能量代谢程度也随之增加。小动物PET/CT能有效检测小鼠体内肩胛处BAT对18F-FDG的代谢情况。 Objective To investigato the effect of different doses of Ephedrine(Eph) and temperature on 18F-FDG uptake in brown adipose tissue (BAT) of Kungming mice using small-animal PET/CT. Methods: 18F-FDG or a mixture of 18F-FDG and Eph (40 mg/kg, 80 mg/kg or 160 mg/kg of body weight) were injected into the peritoneal cavity of mice at different temperatures. One hour after injection, the BAT activity was assessed by measuring SUV with Micro PET/CT. ResultsIn mice without ingection of Eph, the SUV of 18F-FDG, was 1.67 ±0.49 at 25℃, and 2.7±0.26 at 15℃, respectively. In mice injected with 40 mg/kg of Eph, the SUV was 2.20 ±0.36 at 25 ℃, and 5.97 ± 0.21 at 15 ℃, respectively. At 25 ℃, mice injected with 40 mg/kg, 80 mg/kg and 160 mg/kg of Eph, the The SUV of 18F-FDG was 2.20± 0.36, 3.33 ±0.31 and 6.73±0.50 respectively. ConclusionThe cold exposure could cause a greater increase in 18F-FDG uptake in BAT, and the metabolism level of BAT was will increase with the increased dose off. Small-animal PET/C is suitable for the study of 18F-FDG metabolism in interscapular BAT of mice.
