摘要
目的:探讨电针百会和足三里穴对脑缺血再灌注损伤大鼠保护作用是否能通过microRNA途径调节实现,进一步探讨针刺百会、足三里穴对脑缺血再灌注损伤大鼠保护作用的机制。方法:SD大鼠随机分为假手术组、模型组、电针组和药物组。后3组造模后根据再灌注时间又分为1天、3天、5天和7天组。电针组电针百会穴和左侧足三里穴。药物组腹腔注射依达拉奉3 mg/kg。规定时间点取大鼠脑组织和血清,以q PCR法检测脑组织和血清中miRNA-29、miRNA-320表达。结果:同期各个时间点,电针组和药物组脑组织及血清miRNA-29表达低于模型组(P<0.05),电针组与药物组都能下调miRNA-29表达,除血清7天时间点,电针组miRNA-29在同一时间点的表达高于药物组(P<0.05)。同期各个时间点,电针组和药物组脑组织及血清miRNA-320表达高于模型组(P<0.05),电针组与药物组都能上调miRNA-320表达,电针组miRNA-320在同一时间点的表达低于药物组(P<0.05)。结论:针刺大鼠百会、足三里穴能下调脑缺血大鼠脑组织及血清miRNA-29的表达,上调miRNA-320表达,减轻缺血再灌注后脑损伤,针刺对脑缺血再灌注后脑组织有一定的保护作用。
Objective:This study evaluated the hypothesis that acupuncture at GV20 and ST36 could exert neu- roprotective actions in a rat model with middle cerebral artery occlusion (MCAO) by modulating microRNA. Methods:Adult Sprague -Dawley rats were randomly divided into following groups: the sham (S) group, the middle cerebral artery occlusion (MCAO) model groups of 2 h MCAO followed by 1, 3, 5, 7 d of reperfusion, the electro - acupuncture (EA) groups of 2 h MCAO followed by 1,3, 5, 7 d of reperfusion and the edaravone (ED) groups of 2 h MCAO followed by 1, 3, 5, 7 d of reperfusion. The MCAO model was established by placement of a suture to block the middle carotid artery, and reperfusion was triggered by suture removal in all groups except group S. The rats in EA groups received electro - acupuncture treatment by needling at GV20 and left ST36. Rats in groups EA and ED received electroacupuncture and edaravone, respectively, every day. The levels of miRNA - 29 and miRNA - 320 in brain and serum were evaluated by Real - time PCR. Results: Com- pared with the MCAO groups, the levels of miRNA -29c in the brain and serum in the EA groups and ED groups significantly decreased at the same time point(P 〈 0. 05 ) ; compared with the ED groups, the levels of miRNA- 29 in the brain and serum in the EA groups significantly with the MCAO groups, the levels of miRNA - 320 in the brain and increased at 1 d, 3 d and 5 d. Compared serum in the EA groups and ED groups sig- nificantly increased at the same time point( P 〈 0. 05 ) ; the levels of miRNA -320 in the brain and serum in the EA groups were significantly decreased compared with those in the ED groups at the same time point. Conclu- sion:Acupuncture at GV20 and ST36 can decrease miRNA- 29 and increase the level of miRNA- 320 in the brain and serum of rat models with MCAO. Acupuncture at GV20 and ST36 has a neuroproteetive effect on the rat model with MCAO.
