摘要
目的:观察夹脊电针对大鼠脊髓继发性损伤后c-fos mRNA及BNIP3 mRNA表达及其变化规律,以及对CBS运动功能评分的影响,观察夹脊电针对脊髓损伤后神经细胞的保护作用及凋亡机制。方法:用改良Alle’s重物坠落法制备大鼠脊髓损伤后模型,在各时间点观察各组不同时间脊髓内c-fos mRNA及BNIP3 mRNA表达水平以及CBS评分的变化。结果:CBS评分结果显示:7天后,夹脊电针组大鼠的动作完成明显优于假手术组和模型组,具有显著性差异。c-fos mRNA阳性细胞数量表达结果显示:假手术组脊髓内存在少量c-fos mRNA弱阳性的神经细胞,且3个时间点表达的变化不显著;模型组和夹脊电针组的c-fos mRNA表达,在术后均迅速升高,在术后第1天时明显升高,第3天时,表达逐渐下降,第7天时有所回升;夹脊电针组和模型组在各时间点的c-fos mRNA表达阳性细胞数量,与假手术组相比差异具有显著意义(P<0.05);术后第1天时,夹脊电针组与模型组相比,c-fos mRNA表达阳性细胞数低于模型组;术后第3天时,夹脊电针组与模型组相比,不具有显著差异;术后第7天时,夹脊电针组与模型组相比,c-fos mRNA表达阳性细胞数高于模型组。BNIP3 mRNA表达结果显示:夹脊电针组内比较,治疗7天组与治疗3天组相比,BNIP3 mRNA表达水平明显降低(P<0.05),差异具有统计学意义;夹脊电针组与模型对照组相比,治疗7天时,BNIP3 mRNA表达水平有明显降低(P<0.05),具有统计学意义。结论:夹脊电针可促进脊髓损伤后肢体运动、感觉功能的恢复,并通过影响c-fos mRNA及BNIP3 mRNA的表达,促进脊髓损伤后功能的恢复及抑制神经细胞的凋亡。
Objective:To probe into the effect and the change rule of Jiaji electro - acupuncture in the treat- ment of injured spinal cord by observing the number of positive cells of c - fos gene and NIP3 mRNA , and the impact on the CBS movement function score. To further explore the protective mechanism of Jiaji electro - acu- puncture on nerve cells for injured spinal cord and the action of c - fos gene and NIP3 mRNA. Methods:45 healthy male Wistar rats were divided into 3 groups :a Jiaji electro - acupuncture group( n = 15 ), a model control group (n = 15 ), a sham operation group( n = 15). Lamina of vertebra was cut without spinal cord injury in the sham - operation group. Injury of T10 spinal cord was induced by modified Alien's method in other two groups. Observe the expressions of c - fos mRNA and NIP3 mRNA. The expressions of c - fos mRNA and NIP3 mRNA,the changes of CBS movement function score in each group were observed for 1,3 and 7days after spinal injury. Results: The CBS movement function score showed that after 7 days, the complete action of Jiaji electro - acu- puncture group was better than the model control group and sham operation group. The result of the number of positive cells of c - los gene showed that there was a small amount of c - los mRNA weakly positive neurons in sham operation group and had no significant difference at 3 time points. The expression of mRNA of c - los gene increased in postoperative in Jiaji electro -acupuncture group cantly on the first day after operation. The expression graduall seventh day;on the first day after operation, the expression and the model control group. It increased signifi- y declined on the third day and picked up on the of mRNA of c - fos gene of Jiaji electro - acupunc- ture group was lower than that in the model group ( P 〈 0.05 ) ; on the third day, there was no significant differ- ence between Jiaji electro -acupuncture group and model group;on the seventh day, the expression of mRNA of c - fos gene of Jiaji electro - acupuncture group was higher than that in the model group ( P 〈 0. 05 ). The result of NIP3 gene showed that as to the acupuncture in the treatment group, 7 d treatment group and 3 d group were compared and the BNIP3 mRNA expression levels decreased obviously ( P 〈 0. 05 ) ; on the seventy day after operation, the expression of mRNA of BNIP3 gene of Jiaji electro - acupuncture group was lower than that in the model group ( P 〈 O. 05 ). Conclusion: The motor function and sensory function of rats with acute spinal cord in- juries can be improved by Jiaji electro -acupuncture. C -fos mRNA and BNIP3 mRNA were impacted by Jiaji electro - acupuncture, suggesting that c - fos mRNA can promote the recovery of function and inhibition of nerve cell apoptosis of the rats with acute spinal cord injuries.
