结直肠癌粪便miRNA-21的表达及其临床意义 被引量:3

摘要 目的初步探讨粪便miRNA-21在结直肠癌中表达的作用及临床意义。方法收集2014年5月至2014年7月间广西医科大学结直肠肛门外科收治病理诊断明确的30例结直肠癌患者粪便标本,另采集10例健康志愿者粪便作为对照组。采用免疫磁珠法分离粪便脱落细胞,RT-qPCR方法检测粪便miRNA-21的表达,分析miRNA-21的表达水平与临床病理因素的关系。结果 RT-qPCR结果显示在结直肠癌患者粪便miRNA-21的表达水平高于正常人,两者间比较差异有统计学意义,P<0.05;根据临床病理因素的不同对结直肠癌患者进行分组比较,结果显示:Ⅲ+Ⅳ期结直肠癌患者miRNA-21的表达水平要高于Ⅰ+Ⅱ期结直肠癌患者,P<0.05;低分化患者高于中高分化者P<0.05;有淋巴结转移患者明显高于无淋巴结转移者,P<0.05。结论结直肠癌粪便miRNA-21表达的升高可能参与了肿瘤的发生、发展,粪便miRNA-21可能成为结直肠癌潜在的肿瘤标记物,为结直肠癌的早期筛查、诊断提供了新的思路。 Objective To investigate the expression of fecal microRNA-21(miRNA-21)in normal human and patients with colorectal cancer.Methods From May 2014 to July 2014,the fecal of 50 patients with colorectal cancer,who received treatment in the first affiliated hospital of Guangxi Medical Uuniversity,department of colorectal surgery,were collectted in this study.The fecal exfoliated cells were isolated by MACS(magnetic cell sorting).RT-qPCR technique was applied to detected the expression of miRNA-21 and analyzed the correlation of miRNA-21 and clinicopathological factors.Results The RT-qPCR result showed that patients with colorectal cancer expressed high levels of miRNA-21 compared with normal human(P〈0.05).The expression level of miRNA-21 in middle-late stage(Ⅲ+Ⅳ)of colorectal cancer tissues was significantly higher than early stage(Ⅰ+Ⅱ)(P〈0.05).The expression level of miRNA-21 in poorly differentiated tumor tissues was significantly higher than middle-high differentiation(P〈0.05).The expression level of miRNA-21 in colorectal cancer with lymph node metastasis was significantly higher than those without lymph node metastasis(P〈0.05).ConclusionIncreased expression of miRNA-21 may be involved in the development of colorectal cancer,and miRNA-21 may be the potential tumor marker of colorectal cancer.It might provide a new way of thinking for colorectal cancer early screening and diagnosis.
