长春花生物碱类化合物液相色谱/四级杆-飞行时间质谱分析 被引量:2

摘要 目的:建立一种长春花生物碱类化合物的液相色谱/四级杆-飞行时间质谱分析方法。方法:本研究采用大孔吸附树脂柱法富集长春花叶中的生物碱类化合物,经Agilent Zorbax Eclipse Plus C18色谱柱(100 mm×3 mm,1.8μm),乙腈-0.1%甲酸水溶液为流动相进行梯度洗脱,对长春花生物碱类化合物进行分离。结果:采用正离子电喷雾-飞行时间质谱法测定精确分子量,并结合标准品对照快速鉴定出15个生物碱类化合物。结论:该方法结合了液相色谱的高分离效能与高分辨质谱的准确性及选择性,具有快速、稳定可靠的特点,可为长春花细胞培养工艺优化和质量控制提供参考。 Objective: The study is aimed to identify the alkaloids of Catharanthus roseu by LC/Q-TOF MS. Methods: An Agilent Zorbax Eclipse Plus C18 ( 100 mm × 3 mm, 1.8 μm) column was used for the separation of the related substances with a mixture of acetonitrile and 0. 1% formic acid buffer solution as the mobile phase by gradient elution. Results: The alkaloids were speculated by electrospray positive ionization LC-TOF/MS accurate ion mass, and verified further through standard substances determination. Alkaloids were separated under the established HPLC condition. Fifteen related alkaloids were characterized. Conclusion : The LC-MS method is useful for the identification of the alkaloids of C. roseu and the results obtained are valuable for its manufacturing process and quality control.
