摘要
目的:通过生物信息学方法预测得到变形链球菌基因组中可能存在的非编码RNA,并进行序列鉴定。方法:通过4种生物信息学预测细菌非编码RNA的方法,系统分析变形链球菌UA159基因组,对部分序列采用RT-PCR实验进行初步验证,将实验证实的序列进行家族鉴定。结果:预测得到变形链球菌UA159中的s RNA共334条,对其中40条至少由两种生物信息学方法预测得到的序列,用RT-PCR进行初步验证,检测到5条s RNA。经生物信息学鉴定,2条属于已知的家族序列,3条未检测到家族序列。结论:生物信息学方法在预测和鉴定变形链球菌s RNA方面,有重要的应用价值。
Objective: To predict and identify the non-coding RNA sequences in Streptococcus mutans by bioinformatic methods. Methods: The genome of S. mutans UA159 was analyzed with 4 bioinformatic methods for non-coding RNA sequences. Part of sequences was detected by RT-PCR and the family sequences were further identified. Results: Based on the bioinformatic prediction, the total number of non-coding RNAs was 334. 40 of them, predicted by at least 2bioinformatic methods, were verified by RT-PCR. 5 non-coding RNAs were detected. Via bioinformatics identification,Two of them were found belonging to the known family sequences, while the other 3 were not identified. Conclusion:Bioinformatics methods has important applicative value in the prediction and identification of Streptococcus mutans non-coding RNAs.
