金荞麦提取物干预后COPD大鼠模型血清指标及肺组织纤维化指标的检测 被引量:9

摘要 目的:研究金荞麦提取物对慢性阻塞性肺疾病(COPD)大鼠模型血清指标及肺组织纤维化指标的影响。方法:选择成年雄性SD大鼠,采用烟熏法制作COPD模型,给予金荞麦提取物进行治疗。治疗后检测肺纤维化宏观指标、分子指标以及血清中炎症反应相关分子。结果:(1)肺组织纤维化:与对照组比较,模型组的气道阻力、胸腔内压力以及I型胶原(Col I)、III型胶原(Col III)、转化生长因子-β(TGF-β)、Smad3含量增加,肺组织动态顺应性以及水通道蛋白1(AQP1)、水通道蛋白5(AQP5)含量降低;与模型组比较,治疗组的气道阻力、胸腔内压力以及Col I、Col III、TGF-β、Smad3含量降低,肺组织动态顺应性以及AQP1、AQP5含量增加;(2)血清指标:与对照组比较,模型组血清中γ-干扰素(INF-γ)、CXCR3、白介素-17(IL-17)、降钙素原(PCT)、白三烯B4(LTB4)的含量明显升高;与模型组比较,治疗组血清中INF-γ、CXCR3、IL-17、PCT、LTB4的含量明显降低。结论:金荞麦提取物干预能够改善肺组织纤维化、缓解炎症反应程度,是治疗COPD的理想药物。 Objective: To study the effects of Fagopyrum dibotrys extracts on serum markers and pulmonary fibrosis indexes in COPD rat model. Methods: Adult male SD rats were selected, COPD models were made by smoking method. After treatment of Fagopyrum dibotrys extracts, macroeconomic indicators and molecular markers of pulmonary fibrosis as well as serum inflammation related molecules were detected. Results: (1) pulmonary fibrosis: Compared with control group, airway resistance, intrathoracic pressure as well as Col Ⅰ, Col Ⅲ, TGF-β and Smad3 contents of model group increased, and dynamic lung compliance as well as AQP1 and AQP5 contents decreased. Compared with model group, airway resistance, intrathoracic pressure as well as Col I, Col III, TGFJ3 and Smad3 contents of treatment group decreased, and dynamic lung compliance as well as AQP1 and AQP5 contents increased; (2) Serum indicators= Compared with control group, serum INF-γ, CXCR3, IL 17, PCT and LTB4 contents of model group significantly increased; compared with model group, serum INF-γ, CXCR3, IL- 17, PCT and LTB4 contents of treatment group significantly decreased. Oonclusion: Fagopyrum dibotrys extract intervention can improve pulmonary fibrosis and relieve inflammation degree; it's an ideal drug for the treatment of COPD.
