摘要
目的:研究不同生物活性胶原支架对人牙髓细胞生长能力的影响及可能的分子机制。方法:分离培养人牙髓细胞,接种在胶原支架材料(COL)、胶原-生物活性玻璃支架材料(COL-BG)、胶原-生物活性玻璃-聚己内酯支架材料(COL-BG-PCL),检测细胞生长能力及相关基因的表达。结果:COL-BGPCL组hDPSCs的吸光值、迁移率以及上清液中ALP含量均高于COL组、COL-BG组;COL-BG-PCL组的Oct4A、NICD、HMGB1、SDF1、DSPP、DMP-1含量均高于COL组和COL-BG组。结论:COLBG-PCL复合材料有助于促进hDPSCs的增殖、迁移和分化过程,可能的分子机制是增加Oct4A、NICD、HMGB1、SDF1、DSPP、DMP-1的表达。
Objective: To study the effect of different biological activity collagen scaffolds on growth ability of human dental pulp cells and its possible molecular mechanisms. Methods: Human dental pulp cells were isolated and cultured, seeded on collagen scaffold (COL), collagen- bioactive glass scaffolds (COL-BG) and collagen - bioactive glass - PCL scaffolds (COL- BG-PCL). Then growth ability of the cells and expression of related genes were detected. Results: hDPSCs absorbance value, migration rate as well as ALP content in supernatant of COL-BG-PCL group were all higher than those of COL group and COL-BG group; Oct4A, NICD, HMGB1, SDF1, DSPP and DMP-1 contents of COL-BG-PCL group were all higher than those of COL group and COL-BG group. Conclusion: COL-BG-PCL composites contribute to proliferation, migration and dif- ferentiation of hDPSCs; the possible molecular mechanisms are increasing Oct4A, NICD, HMGB1, SDF1, DSPP and DMP-1 expression.
