摘要
环氧合酶(cyclooxygenase,COX)是生物体内参与催化前列腺素生成的一种关键酶,虽然COX有COX-1和COX-2两种同工酶的形式,但这两种同工酶催化底物、抑制剂、组织和细胞内定位以及生理功能有较大差异。本研究利用小鼠生理性孕酮撤退月经模型,通过免疫组化的方法研究COX-1和COX-2在月经期子宫内膜崩解过程中的定位。免疫组化结果显示,两种蛋白质都定位于子宫腔上皮及其附近和内膜崩解区域,且COX-2在子宫内膜蜕膜化区域与基底层分离的外周也有表达,提示COX在参与子宫内膜崩解出血起到一定的作用。
Cyclooxygenase(cox) is the key limited enzyme that catalyzes arachidonic acid into prostaglandin E2(PGE2). There are two isozyme in vivo, COX-1 and COX-2 with large differences in the catalytic substrate,inhibitor, localization in the tissues and cells and physiological function. In this study, through immunohistochemistry, we test the orientation of COX-1 and COX-2 during the menstrual period for endometrial disintegrating by using a mouse menstruation model of physiologic progesterone withdrawal. The immunohistochemistry suggested the localization of these two proteins in the luminal epithelium as well as its nearby area and the shedding regions.Meanwhile, COX-2 also expressed in the peripheral of the separation regions of endometrial decidualization area and basal layer. It indicated an important role of COX in the endometrial disintegration and bleeding.
