摘要
目的:观察清肠化湿方对实验性结肠炎小鼠白介素6(IL-6)反式信号转导trans-signaling的影响,初步探讨该方治疗溃疡性结肠炎(UC)作用机制是否与调控IL-6 trans-signaling有关。方法:TNBS法制备小鼠结肠炎模型,药物干预后,ELISA法检测可溶性白细胞介素6受体(s IL-6R)含量,实时荧光定量PCR法检测IL-6、糖蛋白130(gp130)m RNA相对表达水平,Western Blot法观察结肠黏膜IL-6、gp130蛋白的表达情况。结果:模型组小鼠s IL-6含量、IL-6及gp130 m RNA和蛋白表达水平较正常小鼠明显增高。清肠化湿方可以降低实验性结肠炎小鼠结肠s IL-6水平(P<0.01),降低IL-6及gp130m RNA和蛋白表达水平(P<0.01)。结论:清肠化湿方对小鼠实验性结肠炎发挥良好抗炎效应,可能与调控IL-6 trans-signaling有关。
This study was aimed to observe the influence of Qing-Chang Hua-Shi Recipe(QHR) on IL-6 transsignaling in experimental colitis mice, in order to initially explore the possible mechanisms of QHR for ulcerative colitis(UC). TNBS/ethanol was used in the establishment of colitis mice model. After intervention of medication, ELISA was used in the detection of soluble Interleukin-6 receptor(s IL-6R). Real-time PCR was used to detect the m RNA expression level of IL-6 and glycoprotein 130(gp130). Western blot was used in the observation of protein expression of IL-6 and gp130 in the colonic mucosa. The results showed that the level of s IL-6R, the m RNA and protein expression of IL-6 and gp130 in the model group were significantly higher than that in the control group. QHR was able to reduce the s IL-6R level(P〈0.01), decreased the m RNA and protein expression of IL-6 and gp130(P〈0.01) in the colon tissues among experimental colitis mice. It was concluded that QHR had good antiinflammatory effects on experimental colitis mice. It might be associated with influencing IL-6 trans-signaling.
