摘要 目的采用响应面法优化岗梅总黄酮的提取工艺。方法考察了乙醇体积分数、提取温度和料液比等因素对提取工艺的影响,并在单因素试验的基础之上,根据Box-Behnken中心组合试验设计原理,采用3因素3水平的响应面分析方法,依据回归分析确定各工艺条件的影响因子,以岗梅总黄酮提取率为响应值作响应面和等高线图。结果在分析各个因素的显著性和交互作用后,得出岗梅总黄酮最佳提取工艺为:乙醇体积分数50%,提取温度92℃,料液比1∶45(g∶m L);在此工艺条件下,总黄酮提取率为6.465 mg/g,与预测值(6.535 mg/g)较为接近。结论响应面法对岗梅总黄酮提取方法有较好的预测性,所得提取工艺方法简单、结果可靠。 Objective Response surface methodology( RSM) was used to optimize the extraction technique of total flavonoids from Radix et Caulis Ilicis asprellae. Methods The effects of ethanol concentration,extraction temperature and the ratio of solid to liquid on extraction technique were studied. Base on the single factor test,an orthogonal test design was developed by Box-Behnken central composite test. Results The optimum conditions for total flavonoids in Radix et Caulis Ilicis asprellae were as follows: ethanol concentration of 50%,extraction temperature of 92 ℃,the ratio of solid to liquid of 1 ∶ 45( g ∶ m L). The maximum extraction yield of total flavonoids was 6. 465 mg / g,which was more close to the predicted value( 6. 535 mg/g). Conclusion The response surface methodology has good prediction of extraction technique of the total flavonoids in Radix et Caulis Ilicis asprellae,which is stable,feasible and simple.
